Claims
- 1. An altered lac repressor protein, that:
a) has an increased binding affinity for LacO DNA sequence, or b) has an enhanced inducer responsivity; or c) has both an increased binding affinity for LacO DNA sequence and an enhanced inducer responsivity; wherein the altered lac repressor protein comprises one, or a combination of two or more of the following amino acid changes relative to a natural lac repressor protein: (i) insertion of one or more amino acids, (ii) deletion of one or more amino acids, and (iii) mutation of one or more amino acid residues; and wherein said amino acid changes occur at, or immediately before or after, the positions corresponding to: Arg51, Val52, Ala53, Lys59, Gln60, Ser61, Phe147, Leu148, Asp149, Val150, Ser151, Asp152, Gly187, Pro188, Leu189, Leu295, Leu296, Gly297, Leu319, Pro320, Val321, or Ser322 of the natural lac repressor.
- 2. The altered lac repressor protein of claim 1, comprising one or a combination of two or more of the following mutations: Val52 to Cys or a similar amino acid, Gln60 to Gly or a similar amino acid; Leu148 to Phe, or a similar amino acid; Leu148 to Tyr, or a similar amino acid; Ser151 to Pro, or a similar amino acid; Pro188 to Ser, or a similar amino acid; Leu296 to Ile or a similar amino acid; Leu296 to Met, or a similar amino acid; Pro320 to Ala or a similar amino acid; Pro320 to Gly or a similar amino acid; Val321 to Ile or a similar amino acid, Val321 to Leu, or a similar amino acid; and Val321 to Met or a similar amino acid.
- 3. The altered lac repressor protein of claim 2 which comprises one or a combination of two or more of the following mutations: Val52Cys, Gln60Gly, Leu148Phe, Leu148Tyr, Ser151Pro, Pro188Ser, Leu296Ile, Leu296Met, Pro320Ala, Pro320Gly, Val321Ile, Val321Leu, and Val321Met.
- 4. The altered lac repressor protein of claim 3 which comprises one of the following mutations: Gln60Gly, Leu148Phe, Leu148Tyr, Ser151Pro, Pro188Ser, Leu296Ile, Leu296Met, Pro320Ala, Pro320Gly, Val321Ile, Val321Leu, and Val321Met.
- 5. The altered lac repressor protein of claim 3 that comprises the Gln60Gly mutation.
- 6. The altered lac repressor protein of claim 5 that comprises amino acids 1 to 60 of SEQ ID NO:2.
- 7. The altered lac repressor protein of claim 6 that has the sequence of SEQ ID NO:2.
- 8. The altered lac repressor protein of claim 5 further comprising one or more of the following mutations: Leu148Phe, Leu148Tyr, Ser151Pro, Pro188Ser, Leu296Ile, Leu296Met, Pro320Ala, Pro320Gly, Val321Ile, Val321Leu, and Val321Met.
- 9. The altered lac repressor protein of claim 5 that further compromises the Leu148Phe mutation and an Asp274Asn mutation.
- 10. The altered lac repressor protein of claim 5 that further comprises the Leu148Phe mutation.
- 11. The altered lac repressor of claim 3 that comprises the Leu148Phe mutation.
- 12. The altered lac repressor protein of claim 11 that further comprising a Asp274Asn mutation.
- 13. The altered lac repressor protein of claim 1 that binds LacO DNA with increased affinity relative to wild-type lac repressor protein.
- 14. The altered lac repressor protein of claim 1 that responds to inducer ligand at comparable or lower concentrations of inducer ligand than does the wild-type lac repressor protein.
- 15. The altered lac repressor protein of claim 14 that responds to inducer ligand at lower concentrations of inducer ligand than does the wild-type lac repressor protein.
- 16. The altered lac repressor protein of claim 15 that binds LacO DNA sequence with an increased affinity as compared with the wild-type lac repressor protein.
- 17. The altered lac repressor protein of claim 1 that has an enhanced inducer responsivity as compared with the wild-type lac repressor protein.
- 18. The altered lac repressor protein of claim 17 that binds LacO DNA sequence with an increased affinity as compared with the wild-type lac repressor protein.
- 19. The altered lac repressor protein of claim 1, wherein the ligand binding domain is from an arabinose binding protein.
- 20. The altered lac repressor protein of claim 1, wherein the ligand binding domain is from a ribose binding protein.
- 21. The altered lac repressor protein of claim 1, wherein the ligand binding domain is from a galactose/glucose binding protein.
- 22. A bacterium or eukaryotic cell, comprising DNA encoding the repressor protein of claim 1.
- 23. The bacterium of claim 22 which is Escherichia coli.
