Claims
- 1. A method of transporting a polypeptide into the nucleus of a cell, said method comprising:
expressing in a cell, a phytochrome comprising said polypeptide attached to the apoprotein component of said phytochrome; and exposing said cell to light whereby said phytochrome migrates from the cytoplasm of said cell into the cell nucleus thereby transporting said polypeptide into the nucleus of said cell.
- 2. The method of claim 1, wherein said cell is a cell whose wild-type does not comprises a phtyochrome.
- 3. The method of claim 1, wherein said cell is selected from the group consisting of a bacterial cell, a yeast cell, an insect cell, and a mammalian cell.
- 4. The method of claim 1, wherein said polypeptide is attached to the carboxyl terminus of said apoprotein.
- 5. The method of claim 4, wherein said polypeptide is attached to the carboxyl terminus of said apoprotein with a peptide linker.
- 6. A method of regulating the transcription of a gene in response to light, said method comprising:
expressing, in a cell, a phytochrome comprising a heterologous transactivator or repressor attached to the apoprotein component of said phytochrome; and exposing said cell to light whereby said phytochrome migrates from the cytoplasm of said cell into the cell nucleus and said transactivator or said repressor alters expression of a gene in said nucleus.
- 7. The method of claim 6, wherein said cell is a cell whose wild-type does not comprises a phtyochrome.
- 8. The method of claim 7, wherein said cell is selected from the group consisting of a bacterial cell, a yeast cell, an insect cell, and a mammalian cell.
- 9. The method of claim 6, wherein said transactivator or repressor is attached to a DNA binding protein.
- 10. The method of claim 6, wherein said transactivator or repressor is attached to the carboxyl terminus of the apoprotein comprising said apophytochrome.
- 11. The method of claim 6, wherein said transactivator is selected from the group consisting of VP16, SP1, NF1, GAL4.
- 12. The method of claim 6, wherein said repressor is selected from the group consisting of lex, lac, and tet.
- 13. The method of claim 10, wherein said DNA binding domain is selected from the group consisting of Gal4, and a nuclear receptor DNA binding domain.
- 14. The method of claim 1, wherein said gene is integrated into the genome of said cell.
