Claims
- 1. A method for preparing a viral extract comprising HIV-2 RNA comprising:a) providing a biological sample that comprises cells comprising CD4 positive lymphocytes infected with HIV-2 virus; b) separating the HIV-2 virus from the cells; c) lysing the virus; and d) obtaining a viral extract comprising viral HIV-2 RNA.
- 2. The method of claim 1, wherein the HIV-2 virus is separated from the cells by centrifuging the biological sample and obtaining the virus from the supernatant.
- 3. The method of claim 1, further comprising concentrating the virus.
- 4. The method of claim 3, wherein the virus is concentrated by centrifuging at 60,000 g for 90 minutes.
- 5. The method of claim 1, wherein the virus is lysed with a buffer containing at least one detergent.
- 6. The method of claim 5, wherein the detergent is at least one of Triton X-100, sodium deoxycholate, and SDS.
- 7. The method of claim 5, wherein the buffer comprises 50 mM Tris-HCl pH 7.5, 150 mM NaCl, 1 mM EDTA, 1% Triton X100, 0.1% sodium deoxycholate, and 0.1% SDS.
- 8. The method of claim 5, wherein the buffer comprises 10 mM Tris-HCl pH 7.5, 10 mM NaCl, 1 mM EDTA, 0.5% SDS.
- 9. The method of claim 1, further comprising detecting the HIV-2 RNA.
- 10. The method of claim 9, wherein the HIV-2 RNA is detected using hybridization to a labeled probe.
- 11. The method of claim 10, wherein hybridization is conducted in the following conditions: 6×SSC, 5×Denhart, 50% formamide, for 18 hours at 42° C.
- 12. The method of claim 10, wherein the hybridization step is followed by a washing step.
- 13. The method of claim 12, wherein the washing step is conducted in the following conditions: 0.1×SSC, 0.1% SDS at 65° C.
- 14. The method of claim 9, wherein the HIV-2 RNA is radiolabeled.
- 15. The method of claim 14, wherein the HIV-2 RNA is radiolabeled with 32P.
