Claims
- 1. A method of determining the fertilizing potential of sperm comprising:
- labeling a polypeptide with a light-sensitive moiety, said polypeptide having a sequence comprising a truncated portion of a prosaposin molecule, said truncated portion containing at least a portion of the intervening sequence between the last cystine in saposin A and the first cystine in saposin B therein, wherein said portion of the intervening sequence has at least one active site which binds to sperm by way of a sperm plasma membrane thereon;
- contacting a sample of sperm with said labeled polypeptide;
- exposing said sperm to a light source; and
- measuring the amount of labeled polypeptide per sperm or sperm sample which reacts to said light source.
- 2. The method of claim 1 wherein said moiety is a fluorescent molecule having a characteristic emission wavelength and intensity when excited by light of a certain wavelength.
- 3. A method of determining sperm fertilizing potential comprising:
- producing a first antibody against at least one epitope of a polypeptide exposed while said polypeptide is bound by sperm, said polypeptide having sequence comprising a truncated portion of a prosaposin molecule, said truncated portion containing at least a portion of the intervening sequence between the last cystine in saposin A and the first cystine in saposin B therein, wherein said portion of the intervening sequence has at least one active site which binds to sperm by way of a sperm plasma membrane thereon;
- binding said first antibody to said epitopes;
- binding a second antibody to said first antibody, wherein said second antibody is labeled with a light-sensitive moiety; and
- measuring the amount of labeled second antibody per sperm or sperm sample which reacts to a light source.
- 4. The method of claim 3 wherein the antibody bound by sperm is a Fab-fragment of said antibody labeled with a light-sensitive moiety which reacts to a light source.
- 5. A method for determining the number of sperm binding sites on an egg investment comprising:
- producing a first antibody against at least one epitope of a polypeptide involved in binding the polypeptide to said egg investment, said polypeptide having a sequence comprising a truncated portion of a prosaposin molecule, said truncated portion containing at least a portion of the intervening sequence between the last cystine in saposin A and the first cystine in saposin B therein, wherein said portion of the intervening sequence has at least one active site which binds to sperm by way of a sperm plasma membrane thereon;
- binding said first antibody to said epitopes on said egg investment in vitro;
- binding a second antibody to said first antibody, wherein said second antibody is labeled with a light-sensitive moiety; and
- measuring the amount of labeled second antibody per sperm or sperm sample which reacts to a light source.
- 6. The method of claim 5 wherein the antibody bound by an egg investment is a Fab-fragment of said antibody labeled with a light-sensitive moiety which reacts to a light source.
