Claims
- 1. An isolated nucleic acid molecule comprising a nucleotide sequence that encodes a protein from a Pyrilidae species, wherein said nucleotide sequence is selected from the group consisting of:
a) a nucleotide sequence comprising a sequence encoding an ultraspiracle receptor; b) a nucleotide sequence comprising the sequence set forth in SEQ ID NO:3; c) a nucleotide sequence encoding a polypeptide comprising the amino acid sequence set forth in SEQ ID NO:4.
- 2. An isolated nucleic acid molecule comprising a nucleotide sequence having at least 85% sequence identity to the sequence of SEQ ID NO:3 or to a nucleotide sequence comprising nucleotide 224-1603 of SEQ ID NO:3, wherein said nucleotide sequence encodes a polypeptide having ultraspiracle receptor activity.
- 3. An isolated nucleic acid molecule that encodes a chimeric protein having ultraspiracle receptor activity, wherein said chimeric protein comprises a ligand-binding domain, a DNA-binding domain, and a transactivation domain, and wherein the nucleotide sequence encoding at least one of said domains is set forth in SEQ ID NO:3.
- 4. An expression cassette comprising a promoter operably linked to a nucleotide sequence encoding an ultraspiracle receptor, wherein said nucleotide sequence is selected from the group consisting of:
a) a nucleotide sequence comprising the sequence set forth in SEQ ID NO: 3; b) a nucleotide sequence encoding a polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 4.
- 5. An expression cassette comprising a promoter operably linked to a nucleotide sequence having at least 85% sequence identity to the sequence of SEQ ID NO:3 or to a nucleotide sequence comprising nucleotides 224-1603 of SEQ ID NO:3, wherein said nucleotide sequence encodes a polypeptide having ultraspiracle receptor activity.
- 6. A plant and its parts having stably incorporated in its genome the expression vector of claim 4.
- 7. The plant and its parts of claim 6, wherein said plant further comprises a second expression cassette stably incorporated in its genome, said second expression cassette comprising a transcriptional regulatory region operably linked to a nucleotide sequence encoding a protein of interest, wherein said transcriptional regulatory region is activated by a ligand-receptor complex.
- 8. A plant and its parts having stably incorporated in its genome an expression cassette comprising a promoter operably linked to a nucleotide sequence having at least 85% sequence identity to the sequence of SEQ ID NO:3 or to a nucleotide sequence comprising nucleotides 224-1603 of SEQ ID NO:3, wherein said nucleotide sequence encodes a polypeptide having ultraspiracle receptor activity.
- 9. Transformed seed of the plant of claim 6.
- 10. A plant cell having stably incorporated in its genome the expression cassette of claim 4.
- 11. The plant cell of claim 10, wherein said plant cell further comprises a second expression cassette stably incorporated in its genome, said second expression cassette comprising a transcriptional regulatory region operably linked to a nucleotide sequence encoding a protein of interest, wherein said transcriptional regulatory region is activated by a ligand-receptor complex.
- 12. A plant cell having stably incorporated in its genome an expression cassette comprising a promoter operably linked to a nucleotide sequence having at least 85% sequence identity to the sequence of SEQ ID NO:3 or to a nucleotide sequence comprising nucleotides 224-1603 of SEQ ID NO:3, wherein said sequence encodes a polypeptide having ultraspiracle receptor activity.
- 13. An isolated polypeptide from a Pyrilidae species, wherein said polypeptide is selected from the group consisting of:
a) a polypeptide sequence comprising an ultraspiracle receptor; b) a polypeptide comprising an amino acid sequence set forth in SEQ ID NO: 4; c) a polypeptide encoded by a nucleotide sequence comprising the sequence set forth in SEQ ID NO:3.
- 14. A chimeric protein comprising a ligand-binding domain, a DNA-binding domain, and a transactivation domain, wherein the amino acid sequence of at least one of said domains is set forth in SEQ ID NO:4.
- 15. A method of selectively inducing gene expression of a protein of interest in a plant, said method comprising:
a) stably incorporating into the genome of said plant an expression cassette comprising a promoter operably linked to a first nucleotide sequence encoding an ultraspiracle receptor, wherein said nucleotide sequence is selected from the group consisting of:
i) a nucleotide sequence from a Pyrilidae species comprising a sequence encoding an ultraspiracle receptor; ii) a nucleotide sequence comprising the sequence set forth in SEQ ID NO: 3; iii) a nucleotide sequence encoding a polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 4; b) stably incorporating into the genome of said plant an expression cassette comprising a transcriptional regulatory region operably linked to a second nucleotide sequence encoding said protein of interest, wherein said transcriptional regulatory region is activated by the ligand-receptor complex; c) contacting said plant with a ligand which complexes with said receptor, wherein said receptor-ligand complex interacts with the transcription regulatory region and induces gene expression of the protein of interest.
- 16. A method of selectively inducing gene expression of a protein of interest in a plant, said method comprising the steps of:
a) stably incorporating into the genome of said plant an expression cassette comprising a promoter operably linked to a first nucleic acid sequence encoding an ultraspiracle receptor comprising a ligand-binding domain, a DNA-binding domain, and a transactivation domain, wherein the nucleotide sequence encoding at least one of said domains is selected from the group consisting of:
i) a nucleotide sequence from a Pyrilidae species comprising a sequence encoding an ultraspiracle receptor; ii) a nucleotide sequence set forth in SEQ ID NO:3; iii) a nucleotide sequence encoding a polypeptide comprising an amino acid sequence set forth in SEQ ID NO:4; b) further stably incorporating into the genome of said plant an expression cassette comprising a transcriptional regulatory region operably linked to a second nucleic acid sequence encoding said protein of interest, and wherein said transcriptional regulatory region is activated by the ligand-receptor complex; c) contacting said plant with a ligand which complexes with said receptor, wherein said receptor-ligand complex interacts with the transcription regulatory region and induces gene expression of the protein of interest.
- 17. The method of claim 16, wherein said promoter is tissue-preferred.
- 18. The method of claim 16, wherein said tissue-preferred promoter is selected from the group consisting of anther-preferred, tapetal-preferred, ovule-preferred and pistil-preferred promoters.
- 19. The method of claim 16, wherein expression of said protein of interest disrupts plant fertility.
- 20. The method of claim 16, wherein expression of said protein of interest induces fertility in an otherwise sterile plant.
- 21. The method of claim 16, wherein said plant is a monocot.
- 22. The method of claim 21, wherein said monocot is maize.
