Claims
- 1. An isolated nucleic acid molecule comprising a nucleotide sequence that encodes a protein from a Pyrilidae species, wherein said nucleotide sequence is selected from the group consisting of:
a) a nucleotide sequence comprising a sequence encoding an ecdysone receptor; b) a nucleotide sequence comprising the sequence set forth in SEQ ID NO: 1; c) a nucleotide sequence encoding a polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 2.
- 2. An isolated nucleic acid molecule comprising a nucleotide sequence having at least 85% sequence identity to the sequence set forth in SEQ ID NO:1 or to a sequence comprising nucleotides 368-2005 of SEQ ID NO:1, wherein said nucleotide sequence encodes a polypeptide having ecdysone receptor activity.
- 3. An isolated nucleic acid molecule that encodes a chimeric protein having ecdysone receptor activity, wherein said chimeric protein comprises a ligand-binding domain, a DNA-binding domain, and a transactivation domain, and wherein the nucleotide sequence encoding at least one of said domains is set forth in SEQ ID NO:1.
- 4. An expression cassette comprising a promoter operably linked to a nucleotide sequence encoding an ecdysone receptor, wherein said nucleotide sequence encoding said ecdysone receptor is selected from the group consisting of:
a) a nucleotide sequence from a Pyrilidae species; b) a nucleotide sequence comprising the sequence set forth in SEQ ID NO: 1; c) a nucleotide sequence encoding a polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 2.
- 5. An expression cassette comprising a promoter operably linked to a nucleotide sequence having at least 85% sequence identity to the sequence of SEQ ID NO:1 or to a nucleotide sequence comprising nucleotides 368-2005 of SEQ ID NO:1, wherein said nucleotide sequence encodes a polypeptide having ecdysone receptor activity.
- 6. A plant and its parts having stably incorporated in its genome the expression vector of claim 4.
- 7. The plant and its parts of claim 6, wherein said plant further comprises a second expression cassette stably incorporated in its genome, said second expression cassette comprising a transcriptional regulatory region operably linked to a nucleotide sequence encoding a protein of interest, wherein said transcriptional regulatory region is activated by a ligand-receptor complex.
- 8. A plant and its parts having stably incorporated in its genome an expression cassette comprising a promoter operably linked to a nucleotide sequence having at least 85% sequence identity to the sequence of SEQ ID NO:1 or to a nucleotide sequence comprising nucleotides 368-2005 of SEQ ID NO:1, wherein said nucleotide sequence encodes a polypeptide having ecdysone receptor activity.
- 9. Transformed seed of the plant of claim 8.
- 10. A plant cell having stably incorporated in its genome the expression cassette of claim 4.
- 11. The plant cell of claim 10, wherein said plant cell further comprises a second expression cassette stably incorporated in its genome, said second expression cassette comprising a transcriptional regulatory region operably linked to a nucleotide sequence encoding a protein of interest, wherein said transcriptional regulatory region is activated by a ligand-receptor complex.
- 12. A plant cell having stably incorporated in its genome an expression cassette comprising a promoter operably linked to a nucleotide sequence having at least 85% sequence identity to the sequence of SEQ ID NO:1 or to a nucleotide sequence comprising nucleotides 368-2005 of SEQ ID NO:1, wherein said sequence encodes a polypeptide having ecdysone receptor activity.
- 13. An isolated polypeptide from a Pyrilidae species, wherein said polypeptide is selected from the group consisting of:
a) a polypeptide sequence comprising an ecdysone receptor; b) a polypeptide comprising an amino acid sequence set forth in SEQ ID NO: 2; c) a polypeptide encoded by a nucleotide sequence comprising the sequence set forth in SEQ ID NO: 1.
- 14. A chimeric protein comprising a ligand-binding domain, a DNA-binding domain, and a transactivation domain, wherein the amino acid sequence of at least one of said domains is set forth in SEQ ID NO:2.
- 15. A method of selectively inducing gene expression of a nucleotide sequence of interest in a plant, said method comprising:
a) stably incorporating into the genome of said plant an expression cassette comprising a promoter operably linked to a first nucleotide sequence, wherein said first nucleotide sequence has at least 85% sequence identity to the sequence of SEQ ID NO:1 or to a nucleotide sequence comprising nucleotides 368-2005 of SEQ ID NO:1, and wherein said first nucleotide sequence encodes a polypeptide having ecdysone receptor activity; b) further stably incorporating into the genome of said plant an expression cassette comprising a transcriptional regulatory region operably linked to a nucleotide sequence of interest; and, c) contacting said plant with a ligand which complexes with said receptor to form a receptor-ligand complex, wherein said receptor-ligand complex interacts with said transcription regulatory region and induces gene expression of said nucleotide sequence of interest.
- 16. The method of claim 15, wherein said first nucleotide sequence has at least 95% sequence identity to the sequence of SEQ ID NO:1 or a nucleotide sequence comprising 368-2005 of SEQ ID NO:1.
- 17. A method of selectively inducing gene expression of a protein of interest in a plant, said method comprising the steps of:
a) stably incorporating into the genome of said plant an expression cassette comprising a promoter operably linked to a first nucleic acid sequence encoding a chimeric ecdysone receptor which comprises a ligand-binding domain, a DNA-binding domain, and a transactivation domain, wherein said nucleotide sequence encoding at least one of said domains is encoded by a nucleotide sequence selected from the group consisting of:
i) a nucleotide sequence from a Pyrilidae species comprising a sequence encoding an ecdysone receptor; ii) a nucleotide sequence set forth in SEQ ID NO:1; iii) a nucleotide sequence encoding a polypeptide comprising an amino acid sequence set forth in SEQ ID NO: 2; b) stably incorporating into the genome of said plant an expression cassette comprising a transcriptional regulatory region operably linked to a second nucleic acid sequence encoding said protein of interest, and wherein said transcriptional regulatory region is activated by the ligand-receptor complex; c) contacting said plant with a ligand which complexes with said receptor, wherein said receptor-ligand complex interacts with the transcription regulatory region and induces gene expression of the protein of interest.
- 18. The method of claim 17, wherein said chimeric ecdysone receptor comprises a heterologous domain, which is from a transcriptional activator protein.
- 19. The method of claim 18, wherein said heterologous domain is from a transcriptional activator protein selected from the group consisting of: VP16, maize C1, and GAL4.
- 20. The method of claim 17, wherein said chimeric ecdysone receptor further comprises a heterologous domain from a member of the steroid/thyroid hormone superfamily of nuclear receptors.
- 21. The method of claim 20, wherein said heterologous domain is from a member of the steroid/thyroid hormone superfamily of nuclear receptors selected from the group consisting of: retinoic acid receptor, vitamin D receptor, and thyroid hormone receptor.
- 22. The method of claim 17, wherein said promoter is tissue-preferred.
- 23. The method of claim 22, wherein said tissue-preferred promoter is selected from the group consisting of anther-preferred, tapetal-preferred, ovule-preferred and pistil-preferred promoters.
- 24. The method of claim 17, wherein said ligand is selected from the group comprising a non-steroidal ecdysteroid agonist, 20-hydroxyecdysone or an analog thereof.
- 25. The method of claim 17, wherein expression of said protein of interest disrupts plant fertility.
