Claims
- 1. A method of screening animals to determine those more likely to produce desired growth, appetite and fatness comprising:
obtaining a sample of genetic material from said animal; and assaying for the presence of a genotype in said animal which is associated with favorable growth, appetite and fatness, said genotype characterized by the following:
a) a polymorphism in the Ghrelin gene.
- 2. The method of claim 1 wherein said polymorphism results in an amino acid change of the Ghrelin gene or its equivalent as determined by a BLAST comparison.
- 3. The method of claim 1 wherein said polymorphisms are located in intron 1, exon 2, exon 3, intron 3, and 3′-UTR of the Ghrelin gene.
- 4. The method of claim 1 wherein said genotype is characterized by a polymorphism identified by a restriction enzyme site selected from the group consisting of Msp1, ApaI AluI, RsaI, BsrI, and BglII.
- 5. The method of claim 1 wherein said step of assaying is selected from the group consisting of restriction fragment length polymorphism (RFLP) analysis, minisequencing, MALD-TOF, SINE, heteroduplex analysis, single strand conformational polymorphism (SSCP), denaturing gradient gel electrophoresis (DGGE) and temperature gradient gel electrophoresis (TGGE).
- 6. The method of claim 1 wherein said animal is a pig.
- 7. The method of claim 1 further comprising the step of amplifying the amount of Ghrelin gene or a portion thereof which contains said polymorphism.
- 8. The method of claim 7 wherein said amplification includes the steps of:
selecting a forward and a reverse sequence primer capable of amplifying a region of the Ghrelin gene which contains a polymorphic Msp1, ApaI, AluI, RsaI, BsrI, BglII site.
- 9. A method of screening animals to determine those more likely to exhibit improved growth, appetite and fatness traits comprising:
obtaining a biological sample of material from said animal; and assaying for the presence of a genotype in said animal which is associated with improved growth, appetite and fatness traits said genotype characterized by the following:
a) a polymorphism in the Ghrelin gene, said polymorphism resulting in a Msp1, ApaI, AluI, RsaI, BsrI, BglII site.
- 10. The method of claim 9 further comprising the step of amplifying the amount of Ghrelin gene or a portion thereof which contains said polymorphism.
- 11. A nucleotide sequence or allele which encodes upon expression an Ghrelin protein, said nucleotide sequence comprising a variant due to a polymorphic site selected from the group consisting of Msp1, ApaI, AluI, RsaI, BsrI, and BglII.
- 12. A Ghrelin protein according to claim 11.
- 13. A method of genetically typing an animal comprising:
obtaining a sample of genetic material from said animal; and assaying for the presence of a genotype in said animal, wherein said genotype is characterized by a polymorphism in the ghrelin gene.
- 14. The method of claim 13 wherein said genotype is characterized by a polymorphism identified by a restriction enzyme site selected from the group consisting of Msp1, ApaI AluI, RsaI, BsrI, and BglII.
- 15. A method for indirect selection of a polymorphism in a Ghrelin gene associated with the traits of growth, appetite, and fatness comprising:
selecting specific alleles of an alternative DNA marker associated with said Ghrelin gene, wherein said gene is associated with the traits of growth, appetite, and fatness; making an indirect selection of a polymorphism; and establishing linkage between the specific allele of the alternative DNA and alleles of the DNA marker associated with the traits of growth, appetite, and fatness.
