Novel nanoparticles for delivery of active agents

Abstract
Milled nanoparticles comprising a biolgically active agent, at least one biopolymer and a coating containing at least one coating which is a polymer or ligand are produced using milling and coating techniques which have not previously been used for these applications
Description
Claims
  • 1. A method of preparation of coated particles containing an active agent comprising the steps of: a) planetary milling of a biopolymer excipient and an active agent, thenb) coating the product of step a with at least one coating agent chosen among polymers and ligands.
  • 2. The method of claim 1 where in biopolymer excipient is a complex carbohydrate.
  • 3. The method of claim 2 wherein the complex carbohydrate is a starch, cellulose, collagen, lactose or alginate.
  • 4. The method of claim 1 wherein the active agent is an anti-cancer agent.
  • 5. The method of claim 1 wherein the biologically active agent is chosen from among cisplatin and paclitaxel.
  • 6. The method of claim 1 wherein the biologically active agent is a macromolecule.
  • 7. The method of claim 1 wherein the macromolecule is chosen from among proteins and peptides.
  • 8. A composition of matter comprising a nanoparticle containing a biolgically active agent, at least one biopolymer and a coating containing at least one coating which is a polymer or ligand.
  • 9. The composition of claim 8 wherein the coating is a glycol or lactone
  • 10. The composition of claim 8 wherein the biologically active agent is an anti-cancer agent.
  • 11. The composition of claim 10 wherein the active agent is a peptide or protein.
  • 12. The composition of claim 9 wherein the active agent is an IgG protein.
  • 13. The method of claim 1 wherein the coating agent is at least one of polycaprolactone and polyethylene glycol.
  • 14. The composition of claim 8 wherein the coating contains a targeting peptide.
  • 15. The composition of claim 14 wherein the targeting peptide is a cancer targeting peptide.
  • 16. The method of claim 1 wherein, the coating contains at least one polymer conjugated to a targeting protein or peptide.
