Claims
- 1. N.sup.2 -naphthalenesulfonyl-L-arginine esters and amides having the formula ##STR72## or the acid addition salts thereof with a pharmaceutically acceptable acid wherein R is selected from the class consisting of (1) alkoxy, alkenyloxy, alkynyloxy and cycloalkoxy, respectively containing not more than 10 carbon atoms, aralkyloxy of not more than 15 carbon atoms, tetrahydrofurfuryloxy, and alkoxy of not more than 10 carbon atoms substituted with an alkoxy group of not more than 10 carbon atoms, halogen or nitro; (2) ##STR73## wherein R.sub.1 is selected from the class consisting of alkenyl of not more than 10 carbon atoms, and substituted alkyl containing not more than 20 carbon atoms wherein said substituent is a member selected from the class consisting of alkoxy, alkoxycarbonyl, arylcarbamoyl, acyl, acyloxy, N,N-polymethylenecarbamoyl and carboxy; and R.sub.2 is selected from the class consisting of hydrogen, alkyl and alkenyl, respectively containing not more than 10 carbon atoms; and substituted alkyl containing not more than 20 carbon atoms wherein said substituent is a member selected from the class consisting of alkoxy, alkoxycarbonyl, arylcarbamoyl, acyl, acyloxy, N,N-polymethylenecarbamoyl and carboxy; and (3) ##STR74## wherein Z is a divalent group containing up to 20 carbon atoms, which consists of more than one group selected from the class consisting of methylene --CH.sub.2 --, mono-substituted methylene ##STR75## wherein R.sub.3 is selected from the class consisting of alkyl, acyl, alkoxy and alkoxycarbonyl, respectively containing not more than 10 carbon atoms, and carbamoyl; and disubstituted methylene ##STR76## wherein R.sub.4 and R.sub.5 are alkyl groups of not more than 10 carbon atoms, and which may further contain at least one member selected from the class consisting of cycloalkylene of not more than 10 carbon atoms, phenylene ##STR77## which may be arranged in any order and complete the ##STR78## ring together with the said methylene, monosubstituted methylene or disubstituted methylene; with the proviso that Z is not polymethylene of 3-10 carbon atoms or polymethylene of 3-10 carbon atoms substituted by one or two alkyl groups of not more than 10 carbon atoms and R' is a member selected from the class consisting of 5,6,7,8-tetrahydro-1-naphthyl, 5,6,7,8-tetrahydro-2-naphthyl, 1-naphthyl, 2-naphthyl, 1-naphthyl substituted with one member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 10 carbon atoms; and 2-naphthyl substituted with a member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 10 carbon atoms.
- 2. The compound of claim 1, wherein R is selected from the class consisting of alkoxy, aralkoxy, alkenyloxy, alkynyloxy, alkoxyalkoxy and halogenated alkoxy, respectively containing not more than 10 carbon atoms.
- 3. The compound of claim 1, wherein R is ##STR79## wherein R.sub.1 is selected from the class consisting of alkenyl of not more than 10 carbon atoms and substituted alkyl containing not more than 20 carbon atoms wherein said substituent is a member selected from the class consisting of alkoxy, alkoxycarbonyl and acyl; R.sub.2 is selected from the class consisting of hydrogen, alkyl and alkenyl, respectively containing not more than 10 carbon atoms, and substituted alkyl containing not more than 20 carbon atoms, wherein said substituent is a member selected from the class consisting of alkoxy, alkoxy carbonyl and acyl, with the proviso that R.sub.2 is hydrogen or methyl when R.sub.1 is alkoxycarbonylalkyl.
- 4. The compound of claim 1, wherein R is ##STR80## wherein Z' is selected from the divalent group consisting of monosubstituted methylene ##STR81## wherein R.sub.3 is an acyl group of not more than 10 carbon atoms, cycloalkylene of not more than 10 carbon atoms, and phenylene ##STR82## and n plus m is an integer from 1 to 10.
- 5. The compounds of claim 1, wherein R is a member selected from the class consisting of alkoxy, alkenyloxy, cycloalkoxy, halogenated alkoxy, respectively containing not more than 10 carbon atoms; aralkyloxy, of not more than 15 carbon atoms, and alkoxy of not more than 10 carbon atoms substituted with alkoxy of not more than 10 carbon atoms; and R' is a member selected from the class consisting of 5,6,7,8-tetrahydro-1-naphthyl, 5,6,7,8-tetrahydro-2-naphthyl, 1-naphthyl, 2-naphthyl, 1-naphthyl substituted with one member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 10 carbon atoms and 2-naphthyl substituted with one member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 10 carbon atoms.
- 6. The compounds of claim 1, wherein R is a member selected from the class consisting of alkoxy of 1-8 carbon atoms, aralkyloxy of 7-9 carbon atoms, alkenyloxy of 3-6 carbon atoms, cyclohexyloxy, .omega.-alkoxyalkoxy of 2-6 carbon atoms, and .omega.-chloroalkoxy of 2-6 carbon atoms; and R' is a group selected from the class consisting of 1-naphthyl substituted with one member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 5 carbon atoms and 2-naphthyl substituted with one member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 5 carbon atoms.
- 7. A pharmaceutical composition which comprises an antithrombotically effective amount of a compound of claim 1 and a pharmaceutically acceptable adjuvant.
- 8. A method for inhibiting activity and suppressing activation of thrombin in vivo, which comprises administering to a mammal a pharmaceutically effective amount of a compound of claim 1.
