Claims
- 16. (NEW). An isolated nucleic acid comprising a nucleotide sequence selected from the group consisting of:
(a) SEQ ID NO:1; (b) a nucleotide sequence encoding SEQ ID NO:2; (c) nucleotides 595 through 1191 of SEQ ID NO:1; (d) a nucleotide sequence encoding amino acids 199 through 397 of SEQ ID NO:2; (e) a nucleotide sequence encoding a fragment of SEQ ID NO:2, such that a polypeptide with an amino acid sequence consisting of the fragment has proteinase activity; (f) a nucleotide sequence encoding a variant amino acid sequence that is at least 90% identical to SEQ ID NO:2 or to a fragment of SEQ ID NO:2 having proteinase activity, such that a polypeptide consisting of said variant amino acid sequence has proteinase activity; and (g) the nucleotide sequence of a nucleic acid that hybridizes under high stringency in 6×SSC and 50% formamide at 42° C. with washing conditions of 0.2×SSC and 0.1% SDS at 68° C. to a nucleic acid consisting of SEQ ID NO:1 or its complement, wherein said nucleotide sequence encodes an SVPH1-8 variant having proteinase activity.
- 17. (NEW). The isolated nucleic acid of claim 16, wherein the nucleic acid encodes a polypeptide having a molecular weight of approximately 81 kD as determined by SDS-PAGE.
- 18. (NEW). The isolated nucleic acid of claim 16, wherein the nucleic acid encodes a polypeptide having proteinase activity and comprising an amino acid sequence selected from the group consisting of:
(a) amino acids 27 through 397 of SEQ ID NO:2; (b) amino acids 27 through 501 of SEQ ID NO:2; (c) amino acids 27 through 680 of SEQ ID NO:2; (d) amino acids 199 through 397 of SEQ ID NO:2; (e) amino acids 199 through 501 of SEQ ID NO:2; and (f) amino acids 199 through 680 of SEQ ID NO:2.
- 19. (NEW). The isolated nucleic acid of claim 16, wherein the nucleic acid encodes a polypeptide having proteinase activity and comprising an amino acid sequence that is at least 90% identical to amino acids 199 through 397 of SEQ ID NO:2 or to a fragment of SEQ ID NO:2 having proteinase activity.
- 20. (NEW). The isolated nucleic acid of claim 16, wherein the nucleic acid encodes a polypeptide comprising amino acids 199 through 397 of SEQ ID NO:2.
- 21. (NEW). The isolated nucleic acid of claim 16, wherein the nucleic acid encodes a polypeptide consisting of a fragment of SEQ ID NO:2 having proteinase activity.
- 22. (NEW). An isolated nucleic acid encoding a fusion protein and comprising the nucleotide sequence of the nucleic acid of claim 16, wherein said nucleic acid encoding a fusion protein further comprises a nucleotide sequence encoding a polypeptide selected from the group consisting of an Fc polypeptide and a peptide linker.
- 23. (NEW). A recombinant vector comprising the nucleic acid of claim 16 that directs the expression of an SVPH1-8 polypeptide encoded by said nucleic acid.
- 24. (NEW). A host cell transformed, transfected, or transduced with the recombinant vector of claim 23.
- 25. (NEW). A method for the expression of an SVPH1-8 polypeptide having proteinase activity, the method comprising culturing a host cell of claim 24 under conditions promoting expression of said SVPH1-8 polypeptide.
- 26. (NEW). The method of claim 25, further comprising recovering the expressed SVPH1-8 polypeptide.
- 27. (NEW). An isolated nucleic acid comprising a nucleotide sequence selected from the group consisting of:
(a) SEQ ID NO:1; (b) a nucleotide sequence encoding SEQ ID NO:2; (c) nucleotides 1192 through 1503 of SEQ ID NO:1; (d) a nucleotide sequence encoding amino acids 398 through 501 of SEQ ID NO:2; (e) a nucleotide sequence encoding a fragment of SEQ ID NO:2, such that a polypeptide with an amino acid sequence consisting of the fragment has disintegrin activity; (f) a nucleotide sequence encoding a variant amino acid sequence that is at least 90% identical to SEQ ID NO:2 or to a fragment of SEQ ID NO:2 having disintegrin activity, such that a polypeptide consisting of said variant amino acid sequence has disintegrin activity; and (g) the nucleotide sequence of a nucleic acid that hybridizes under high stringency in 6×SSC and 50% formamide at 42° C. with washing conditions of 0.2×SSC and 0.1% SDS at 68° C. to a nucleic acid consisting of SEQ ID NO:1 or its complement, wherein said nucleotide sequence encodes an SVPH1-8 variant having disintegrin activity.
- 28. (NEW). The isolated nucleic acid of claim 27, wherein the nucleic acid encodes a polypeptide having a molecular weight of approximately 81 kD as determined by SDS-PAGE.
- 29. (NEW). The isolated nucleic acid of claim 27, wherein the nucleic acid encodes a polypeptide having disintegrin activity and comprising an amino acid sequence selected from the group consisting of:
(a) amino acids 27 through 501 of SEQ ID NO:2; (b) amino acids 27 through 680 of SEQ ID NO:2; (c) amino acids 199 through 501 of SEQ ID NO:2; (d) amino acids 199 through 680 of SEQ ID NO:2; (e) amino acids 398 through 501 of SEQ ID NO:2; and (f) amino acids 398 through 680 of SEQ ID NO:2.
- 30. (NEW). The isolated nucleic acid of claim 27, wherein the nucleic acid encodes a polypeptide comprising amino acids 398 through 501 of SEQ ID NO:2.
- 31. (NEW). The isolated nucleic acid of claim 27, wherein the nucleic acid encodes a polypeptide consisting of a fragment of SEQ ID NO:2 having disintegrin activity.
- 32. (NEW). An isolated nucleic acid encoding a fusion protein and comprising the nucleotide sequence of the nucleic acid of claim 27, wherein said nucleic acid encoding a fusion protein further comprises a nucleotide sequence encoding a polypeptide selected from the group consisting of an Fc polypeptide and a peptide linker.
- 33. (NEW). A recombinant vector comprising the nucleic acid of claim 27 that directs the expression of an SVPH1-8 polypeptide encoded by said nucleic acid.
- 34. (NEW). A host cell transformed, transfected, or transduced with the recombinant vector of claim 33.
- 35. (NEW). A method for the expression of an SVPH1-8 polypeptide having disintegrin activity, the method comprising culturing a host cell of claim 34 under conditions promoting expression of said SVPH1-8 polypeptide.
- 36. (NEW). The method of claim 35, further comprising recovering the expressed SVPH1-8 polypeptide.
- 37. (NEW). An isolated nucleic acid encoding a polypeptide having disintegrin activity and comprising an amino acid sequence that is at least 90% identical to amino acids 398 through 501 of SEQ ID NO:2 or to a fragment of SEQ ID NO:2 having disintegrin activity.
