Claims
- 1. A method of indicating neurodegenerative disorders characterized by deposition of cerebral amyloid, comprising detecting a decrdease in the ratio of .alpha.-sAPP to sAPP or a decrease in the amount of .alpha.-sAPP in a sample of CSF, wherein .alpha.-sAPP is the processed APP fragment whose generation precludes A.beta. formation and that comprises the N-terminus of APP, but lacks the C-terminus.
- 2. The method of claim 1, wherein the decrease in the ratio or amount is compared to a control.
- 3. The method of claim 2, wherein the control is the ratio or amount in CSF from individuals who do not have this disorder or is a predetermined standard ratio or amount.
- 4. The method of claim 1, wherein the amount of .alpha.-sAPP is quantitated using a sandwich enzyme-linked immunosorbent assay.
- 5. The method of claim 4, wherein:
- a first antibody is attached to a solid support and is directed against an amino terminal epitope of sAPP;
- the first antibody is contacted with a sample of human CSF to form a complex of antibody and any reacting antigens in the CSF, and the complex is then contacted with a second antibody that reacts with the N-terminus of A.beta. present in .alpha.-sAPP;
- the second antibody is an anti-human sAPP antibody conjugated to the enzyme.
- 6. The method of claim 5, wherein the enzyme catalyzes reaction of a chromagenic substrate, whereby cleavage of the chromagenic substrate by the enzyme of the second antibody conjugate results in a detectable color change.
- 7. The method of claim 1, wherein the amount of .alpha.-sAPP is quantitated using an immunoblotting assay and laser densitometry.
- 8. The method of claim 5, wherein the first antibody is a monoclonal antibody.
- 9. The method of claim 8, wherein the first antibody is P2-1.
- 10. The method of claim 5, wherein the second antibody is a polyclonal antibody.
- 11. The method of claim 10, wherein the second antibody is rabbit antibody 3369.
- 12. The method of claim 5, wherein the second antibody is a monoclonal antibody.
- 13. The method of claim 12, wherein the second antibody is 6E10.
- 14. The method of claim 5, wherein the enzyme is horseradish peroxidase.
- 15. The method of claim 1, wherein the amount of .alpha.-sAPP in the sample is compared to the amount of .alpha.-sAPP in a control CSF sample.
- 16. The method of claim 15, wherein the control CSF is from a non-afflicted individual or is a predetermined standard amount.
- 17. The method of claim 1, wherein the amount of .alpha.-sAPP is assessed by isolating total sAPP from the CSF, and then identifying the portion of total sAPP that contains only an amino terminal part of A.beta..
- 18. The method of claim 1, wherein the amount of sAPP is quantitated using a sandwich enzyme-linked immunosorbent assay.
- 19. The method of claim 17, wherein the portion of total sAPP that comprises the amino terminal part of A.beta. is identified using a monoclonal antibody specific therefor.
- 20. The method of claim 17, wherein the antibody is 6E10.
- 21. The method of claim 15, wherein the disorder is Alzheimer's Disease.
- 22. The method of claim 1, wherein total sAPP is the amount of protein in the sample that contains the N-terminus of APP.
