Claims
- 1. A method for obtaining a basic polypeptide factor having a molecular weight of from about 30 kD to about 36 kD on SDS-polyacrylamide gel electrophoresis which has mitogenic activity on rat Schwann cells in the presence of fetal calf plasma and, when isolated using reversed-phase HPLC retains at least 50% of its activity after 10 weeks incubation in 0.1% trifluoroacetic acid at 4.degree. C., comprising extracting vertebrate brain material to obtain protein, subjecting said protein to chromatographic purification comprising hydroxylapatite HPLC and thereafter to SDS-polyacrylamide gel electrophoresis, and collecting a fraction therefrom which has an observed molecular weight of about 30 kD to 36 kD which stimulates division of rat Schwann cells against a background of fetal calf plasma.
- 2. The method of claim 1, wherein the vertebrate brain material is pituitary material.
- 3. The method of claim 2, wherein said pituitary material is bovine pituitary material.
- 4. The method of claim 1, comprising subjecting said vertebrate brain material to carboxymethyl cellulose chromatography prior to hydroxylapatite HPLC.
- 5. The method as claimed in claim 1 further comprising carrying out at least one step selected from the group consisting of cation exchange chromatography, gel filtration and reversed-phase HPLC, following said hydroxylapatite HPLC.
- 6. The method of claim 1 further comprising assaying for mitogenic activity by assaying purified fractions for stimulating the division of rat Schwann cells in the presence of fetal calf plasma following each step.
- 7. A method for obtaining basic polypeptide factor having a molecular weight, under non-reducing conditions, of from about 55 kD to about 63 kD via SDS-polyacrylamide gel electrophoresis, said polypeptide factor having mitogenic activity on rat Schwann cells in the presence of fetal calf plasma, and when isolated using reversed-phase HPLC retains at least about 50% of said mitogenic activity after 4 days incubation in 0.1% trifluoroacetic acid at 4.degree. C., comprising extracting vertebrate brain material to obtain protein, subjecting said protein to chromatographic purification comprising hydroxylapatite HPLC and thereafter to SDS-polyacrylamide gel electrophoresis and collecting a fraction therefrom which has an observed molecular weight of about 55 kD to 63 kD and under non-reducing conditions, and which exhibits mitogenic activity by stimulating the division of rat Schwann cells against a background of fetal calf plasma.
- 8. The method of claim 7, wherein said vertebrate brain material is pituitary material.
- 9. The method of claim 7, wherein said pituitary material is bovine pituitary material.
- 10. The method of claim 7 wherein said comprising subjecting said vertebrate brain material to carboxymethyl cellulose chromatography prior to hydroxylapatite HPLC.
- 11. The method of claim 7, further comprising carrying out at least one step selected from the group consisting of cation exchange chromatography, gel filtration, and reversed-phase HPLC following hydroxylapatite HPLC.
- 12. The method of claim 7, further comprising assessing for mitogenic activity by assaying purified fractions for stimulating the division of rat Schwann cells in the presence of fetal calf plasma following each step.
