Claims
- 1. A Bacillus cell containing a mutation in the RP-I-encoding gene (bpr) of said Bacillus cell, said gene being the same as, or an evolutionary homologue of the RP-I gene of Bacillus subtilis, and said mutation resulting in inhibition of the production by said cell of proteolytically active RP-I gene product.
- 2. The Bacillus cell of claim 1, further containing mutations in the apr and npr genes encoding extracellular proteases, said mutations resulting in inhibition of the production by said cell of proteolytically active apr and npr gene products.
- 3. A Bacillus cell containing a mutation in the RP-II-encoding gene of said Bacillus cell, said gene being the same as, or an evolutionary homologue of the RP-II (mpr) gene of Bacillus subtilis, and said mutation resulting in inhibition of the production by said cell of proteolytically active RP-II gene product.
- 4. The Bacillus cell of claim 1, further containing a mutation in the RP-II-encoding gene of said Bacillus cell, said gene being the same as, or an evolutionary homologue of the RP-II (mpr) gene of Bacillus subtilis, and said mutation resulting in inhibition of the production by said cell of proteolytically active RP-II gene product.
- 5. A Bacillus cell containing a mutation in the epr gene of said Bacillus cell, said gene being the same as, or an evolutionary homologue of the epr gene of Bacillus subtilis, and said mutation resulting in inhibition of the production by said cell of proteolytically active epr gene product, further containing a mutation in the RP-II-encoding gene of said Bacillus cell, said gene being the same as, or an evolutionary homologue of the RP-II (mpr) gene of Bacillus subtilis, and said mutation resulting in inhibition of the production by said cell of proteolytically active RP-II gene product.
- 6. A Bacillus cell containing a mutation in the epr gene of said Bacillus cell, said gene being the same as, or an evolutionary homologue of the epr gene of Bacillus subtilis, and said mutation resulting in inhibition of the production by said cell of proteolytically active epr gene product, said cell further containing a mutation in the RP-I-encoding gene of said Bacillus cell, said gene being the same as, or an evolutionary homolog of the RP-I (bpr) gene of Bacillus subtilis, and said mutation resulting in inhibition of the production by said cell of proteolytically active RP-I gene product.
- 7. The Bacillus cell of claim 2, said cell further containing a mutation in the RP-II-encoding gene (mpr) of said Bacillus Cell, said gene being the same as, or an evolutionary homologue of the RP-II gene of Bacillus subtilis, and said mutation resulting in inhibition of the production by said cell of proteolytically active RP-II gene product.
- 8. The Bacillus cell of claim 6, said cell further containing a mutation in the RP-II-encoding gene (mpr) of said Bacillus cell, said gene being the same as, or an evolutionary homologue of the RP-II gene of Bacillus subtilis, and said mutation resulting in inhibition of the production by said cell of proteolytically active RP-II gene product.
- 9. The Bacillus cell of claim 6, further containing mutations in the apr and npr genes encoding extracellular proteases, said mutations resulting in inhibition of the production by said cell of proteolytically active apr and npr gene products.
- 10. The Bacillus cell of claim 5, further containing mutations in the apr and npr genes encoding extracellular proteases, said mutations resulting in inhibition of the production by said cell of proteolytically active apr and npr gene products.
- 11. The Bacillus cell of claim 3, further containing mutations in the apr and npr genes encoding extracellular proteases, said mutations resulting in inhibition of the production by said cell of proteolytically active apr and npr gene products.
- 12. The Bacillus cell of claim 9, said cell further containing a mutation in the RP-II-encoding gene (mpr) of said Bacillus cell, said gene being the same as, or an evolutionary homologue of the RP-II gene of Bacillus subtilis, and said mutation resulting in inhibition of the production by said cell of proteolytically active RP-II gene product.
- 13. The Bacillus cell of claim 9, each of said mutations comprising a deletion within the coding region of said mutated gene.
- 14. The Bacillus cell of claim 10, each of said mutations comprising a deletion within the coding region of said mutated gene.
- 15. The Bacillus cell of claim 12, each of said mutations comprising a deletion within the coding region of said mutated gene.
- 16. The Bacillus cell of claim 9, said cell further containing a mutation in the isp-1 gene encoding an intracellular protease.
- 17. The Bacillus cell of claim 16, further containing a mutation in the RP-II-encoding gene (mpr) of said Bacillus cell, said gene being the same as, or an evolutionary homologue of the RP-II gene of Bacillus subtilis, and said mutation resulting in inhibition of the production by said cell of proteolytically active RP-II gene product.
- 18. The Bacillus cell of claim 9, said cell further containing a mutation which reduces said cell's capacity to produce one or more sporulation-dependent proteases.
- 19. The Bacillus cell of claim 12, said cell further containing a mutation which reduces said cell's capacity to produce one or more sporulation-dependent proteases.
- 20. The Bacillus cell of claim 18 or 19 wherein said mutation blocks sporulation at an early stage but does not eliminate the cell's ability to be transformed by purified DNA.
- 21. The Bacillus cell of claim 20, said mutation being in the spoOA gene.
- 22. The Bacillus cell of claim 21, said cell being Bacillus subtills.
- 23. The Bacillus cell of any of claims 1-19, further comprising a gene encoding a heterologous polypeptide.
