Claims
- 1. An isolated nucleic acid comprising a nucleic acid segment of interest linked to one or more nucleic acid segments encoding at least two Green Fluorescent Proteins.
- 2. The isolated nucleic acid of claim 1, wherein the nucleic acid segment of interest encodes a polypeptide.
- 3. The isolated nucleic acid of claim 1, comprising two nucleic acid segments each of which encodes a Green Fluorescent Protein one of which is linked at the 3′ and the other of which is linked at the 5′ end of the nucleic acid segment of interest.
- 4. The nucleic acid of claim 1, wherein the Green Fluorescent Proteins are both linked at the 5′ end of the nucleic acid segment of interest.
- 5. The nucleic acid of claim 1, wherein the Green Fluorescent Proteins are both linked at the 3′ end of the nucleic acid segment of interest.
- 6. The nucleic acid of claim 1, wherein the nucleic acid segments each of which encodes a Green Fluorescent Protein both of which are linked to a portion of the nucleic acid segment of interest.
- 7. The nucleic acid of claim 1, wherein the nucleic acid segment of interest is linked to at least three nucleic acid segments each of which encodes a Green Fluorescent Protein.
- 8. The nucleic acid of claim 1, wherein the nucleic acid segment which encodes a Green Fluorescent Protein is a GFP mutant, GFPmut1(pEGFP-N1).
- 9. The nucleic acid of claim 1, comprising two nucleic acid segments each of which encodes a Green Fluorescent Protein wherein the Green Fluorescent Proteins are the same.
- 10. The nucleic acid of claim 1, comprising two nucleic acid segments each of which encodes a Green Fluorescent Protein wherein the Green Fluorescent Proteins are different.
- 11. The isolated nucleic acid of claim 1, wherein the nucleic acid is DNA.
- 12. The isolated nucleic acid of claim 1, wherein the nucleic acid segment of interest is a cytoskeletal protein.
- 13. The isolated nucleic acid of claim 12, wherein the cytoskeletal protein is the full-length Ensconsin (E-MAP-115) or of the microtubule-binding domain of Ensconsin (EMTB).
- 14. A vector comprising the isolated nucleic acid of claim 1.
- 15. The vector of claim 14, wherein the vector is a plasmid.
- 16. A polypeptide encoded by the nucleic acid of claim 1.
- 17. A protein conjugate containing at least two molecules of Green Fluorescent Protein and a protein of interest.
- 18. A method for detecting a nucleic acid segment of interest in a living cell which comprises:
(a) transfecting the living cell with an isolated nucleic acid comprising a nucleic acid which encodes the nucleic acid segment of interest linked to at least two nucleic acids segments each of which encodes a Green Fluorescent Protein; (b) culturing the transfected cell in conditions permitting expression of Green Fluorescent Protein and the nucleic acid segment of interest; and (c) detecting the fluorescence of the Green Fluorescent Protein, thereby detecting the nucleic acid segment of interest in a cell.
- 19. A method for detecting a nucleic acid segment of interest in a living cell which comprises:
(a) transfecting the living cell with an isolated nucleic acid comprising a nucleic acid which encodes the nucleic acid segment of interest and another nucleic acid which encodes at least two molecules of Green Fluorescent Protein; (b) culturing the transfected cell in conditions permitting expression of Green Fluorescent Protein and the nucleic acid segment of interest; and (c) detecting the fluorescence of the Green Fluorescent Protein, thereby detecting the nucleic acid segment of interest, its movement and dynamics in a cell.
- 20. The method for detecting of claim 18, wherein the detection is by fluorescence microscopy or time lapse imaging.
- 21. The method of claim 18, wherein the cell normally expresses the nucleic acid segment of interest.
- 22. The method of claim 18, wherein the number of molecules of the nucleic acid segment of interest detected is between one to ten.
- 23. The nucleic acid of claim 1, wherein the nucleic acid segment which encodes a Green Fluorescent Protein is a GFP mutant.
- 24. The nucleic acid of claim 23, wherein the nucleic acid segment which encodes a Green Fluorescent Protein is a Fluorescent Protein of a color other than green.
