Claims
- 1. A chimeric protein comprising at least a first and a second polypeptide, wherein the first polypeptide comprises Borrelia burgdorferi OspC and wherein the second polypeptide comprises Borrelia burgdorferi OspA, such that OspC comprises the N-terminus of the protein.
- 2. A protein of claim 1, wherein the OspC polypeptide is present in unlipidated form.
- 3. A protein of claim 1, wherein the OspA polypeptide is present in unlipidated form.
- 4. A protein of claim 1, wherein the OspA and OspC polypeptides are from the same strain of Borrelia burgdorferi.
- 5. A protein of claim 1, wherein the OspA and OspC polypeptides are from the same genospecies of Borrelia burgdorferi.
- 6. A protein of claim 1, wherein the OspA and OspC polypeptides are from different strains of Borrelia burgdorferi.
- 7. A protein of claim 1, wherein the OspA and OspC polypeptides are from different genospecies of Borrelia burgdorferi.
- 8. A protein of claim 1, wherein the OspA polypeptide comprises at least a first OspA amino acid sequence from a first strain of Borrelia burgdorferi and a second OspA amino acid sequence from a second strain of Borrelia burgdorferi.
- 9. A protein of claim 8, wherein the first strain of Borrelia burgdorferi is from a first genospecies of Borrelia burgdorferi and wherein the second strain of Borrelia burgdorferi is from a second genospecies of Borrelia burgdorferi.
- 10. A protein of claim 8, wherein the first OspA polypeptide fragment comprises a Borrelia burgdorferi OspA polypeptide from about amino acid residue 18 to about amino acid residue 216, and wherein the second OspA polypeptide fragment comprises a Borrelia burgdorferi OspA polypeptide from about amino acid residue 217 to about amino acid residue 273.
- 11. A protein of claim 8, wherein the first OspA polypeptide fragment comprises a Borrelia burgdorferi OspA polypeptide from about amino acid residue 132 to about amino acid residue 217, and wherein the second OspA polypeptide fragment comprises a Borrelia burgdorferi OspA polypeptide from about amino acid residue 218 to about amino acid residue 273.
- 12. A protein of claim 8, wherein the OspA polypeptide comprises at least four separate OspA polypeptide fragments.
- 13. A protein of claim 12, wherein the first OspA polypeptide fragment comprises a Borrelia burgdorferi OspA polypeptide from about amino acid residue 30 to about amino acid residue 150, wherein the second OspA polypeptide fragment comprises a Borrelia burgdorferi OspA polypeptide from about amino acid residue 151 to about amino acid residue 179, wherein the third OspA polypeptide fragment comprises a Borrelia burgdorferi OspA polypeptide from about amino acid residue 180 to about amino acid residue 216, and wherein the fourth OspA polypeptide fragment comprises a Borrelia burgdorferi OspA polypeptide from about amino acid residue 217 to about amino acid residue 273.
- 14. A protein of claim 1, wherein the first polypepetide comprises a Borrelia burgdorferi OspC polypeptide from about amino acid residue 19 to about amino acid residue 213.
- 15. A protein of claim 1, wherein the first polypepetide comprises a Borrelia burgdorferi OspC polypeptide from about amino acid residue 19 to about amino acid residue 211.
- 16. A protein of claim 1, wherein the first polypeptide comprises a Borrelia burgdorferi OspC polypeptide from about amino acid residue 19 to about amino acid residue 204.
- 17. A protein of claim 1, wherein the second polypeptide comprises a Borrelia burgdorferi OspA polypeptide from about amino acid 18 to about amino acid 273.
- 18. A protein of claim 1, wherein the second polypeptide comprises a Borrelia burgdorferi OspA polypeptide from about amino acid 132 to about amino acid 216.
- 19. A chimeric protein comprising an amino acid sequence selected from the group consisting of: SEQ ID NOs: 146, 148, 150, 152, 154, 156, 158, 160, 162, 164, 166, 168, 170, 172, 174, 176, 178 and 180.
- 20. A nucleic acid encoding a protein comprising at least a first and a second polypeptide, wherein the first polypeptide comprises Borrelia burgdorferi OspC and wherein the second polypeptide comprises Borrelia burgdorferi OspA, such that OspC comprises the N-terminus of the encoded protein.
- 21. A nucleic acid encoding a protein comprising at least a first and a second polypeptide, wherein the first polypeptide comprises a Borrelia burgdorferi OspC polypeptide from about amino acid residue 19 to about amino acid residue 211, and wherein the second polypeptide comprises a Borrelia burgdorferi OspA polypeptide selected from the group consisting of:
(a) a Borrelia burgdorferi OspA polypeptide from about amino acid residue 18 to about amino acid residue 273, (b) a Borrelia burgdorferi OspA polypeptide from about amino acid residue 132 to about amino acid residue 216, (c) a Borrelia burgdorferi polypeptide comprising at least two separate OspA polypeptide fragments, wherein the first OspA polypeptide fragment comprises an OspA polypeptide from about amino acid residue 18 to about amino acid residue 216 and, wherein the second OspA polypeptide comprises an OspA polypeptide from about amino acid residue 217 to about amino acid residue 273, (d) a Borrelia burgdorferi polypeptide comprising at least two separate OspA polypeptide fragments, wherein the first OspA polypeptide fragment comprises an OspA polypeptide from about amino acid residue 132 to about amino acid residue 216 and, wherein the second OspA polypeptide comprises an OspA polypeptide from about amino acid residue 217 to about amino acid residue 273, and (e) a Borrelia burgdorferi polypeptide comprising at least four separate OspA polypeptide fragments, wherein the first OspA polypeptide fragment comprises an OspA polypeptide from about amino acid residue 30 to about amino acid residue 150, wherein the second OspA polypeptide comprises an OspA polypeptide from about amino acid residue 151 to about amino acid residue 179, wherein the third OspA polypeptide comprises an OspA polypeptide from about amino acid residue 181 to about amino acid residue 216 and, wherein the fourth OspA polypeptide comprises an OspA polypeptide from about amino acid residue 216 to about amino acid residue 273.
- 22. A nucleic acid encoding a protein comprising at least a first and a second polypeptide, wherein the first polypeptide comprises a Borrelia burgdorferi OspC polypeptide from about amino acid residue 19 to about amino acid residue 204, and wherein the second polypeptide comprises a Borrelia burgdorferi OspA polypeptide selected from the group consisting of:
(a) a Borrelia burgdorferi OspA polypeptide from about amino acid residue 18 to about amino acid residue 273, (b) a Borrelia burgdorferi OspA polypeptide from about amino acid residue 132 to about amino acid residue 216, (c) a Borrelia burgdorferi polypeptide comprising at least two separate OspA polypeptide fragments, wherein the first OspA polypeptide fragment comprises an OspA polypeptide from about amino acid residue 18 to about amino acid residue 216 and, wherein the second OspA polypeptide comprises an OspA polypeptide from about amino acid residue 217 to about amino acid residue 273, (d) a Borrelia burgdorferi polypeptide comprising at least two separate OspA polypeptide fragments, wherein the first OspA polypeptide fragment comprises an OspA polypeptide from about amino acid residue 132 to about amino acid residue 216 and, wherein the second OspA polypeptide comprises an OspA polypeptide from about amino acid residue 217 to about amino acid residue 273, and (e) a Borrelia burgdorferi polypeptide comprising at least four separate OspA polypeptide fragments, wherein the first OspA polypeptide fragment comprises an OspA polypeptide from about amino acid residue 30 to about amino acid residue 150, wherein the second OspA polypeptide comprises an OspA polypeptide from about amino acid residue 151 to about amino acid residue 179, wherein the third OspA polypeptide comprises an OspA polypeptide from about amino acid residue 181 to about amino acid residue 216 and, wherein the fourth OspA polypeptide comprises an OspA polypeptide from about amino acid residue 216 to about amino acid residue 273.
- 23. A nucleic acid of any one of claims 20, 21 or 22, wherein the first nucleic acid sequence is from a first strain of Borrelia burgdorferi and the second nucleic acid sequence is from a second strain of Borrelia burgdorferi.
- 24. A nucleic acid of any one of claims 20, 21 or 22, wherein the first nucleic acid sequence is from a first genospecies of Borrelia burgdorferi and the second nucleic acid sequence is from a second genospecies of Borrelia burgdorferi.
- 25. A nucleic acid of any one of claims 20, 21 or 22, wherein the first nucleic acid sequence is from a first strain of Borrelia burgdorferi and the second nucleic acid sequence is from a second strain of Borrelia burgdorferi.
- 26. A nucleic acid any one of claims 20, 21 or 22, wherein the first nucleic acid sequence is from a first genospecies of Borrelia burgdorferi and the second nucleic acid sequence is from a second genospecies of Borrelia burgdorferi.
- 27. A nucleic acid selected from the group consisting of: SEQ ID NOs: 145, 147, 149, 151, 153, 155, 157, 159, 161, 163, 165, 167, 169, 171, 173, 175, 177 and 179.
- 28. An expression vector comprising an isolated DNA having a nucleotide sequence selected from the group consisting of: SEQ ID NOs: 145, 147, 149, 151, 153, 155, 157, 159, 161, 163, 165, 167, 169, 171, 173, 175, 177 and 179.
- 29. An expression vector comprising an isolated DNA encoding a polypeptide having an amino acid sequence selected from the group consisting of: SEQ ID NOs: 146, 148, 150, 152, 154, 156, 158, 160, 162, 164, 166, 168, 170, 172, 174, 176, 178 and 180.
- 30. A host cell comprising a recombinant nucleic acid having a nucleotide sequence selected from the group consisting of: SEQ ID NOs: 145, 147, 149, 151, 153, 155, 157, 159, 161, 163, 165, 167, 169, 171, 173, 175, 177 and 179.
- 31. A host cell comprising a recombinant nucleic acid encoding a protein having an amino acid sequence selected from the group consisting of: SEQ ID NOs: 146, 148, 150, 152, 154, 156, 158, 160, 162, 164, 166, 168, 170, 172, 174, 176, 178 and 180.
- 32. A method of producing a chimeric Borrelia burgdorferi protein comprising;
a) selecting a polynucleotide encoding OspC or an antigenic portion thereof; b) selecting a polynucleotide encoding OspA or an antigenic portion thereof; and c) ligating the polynucleotide of a) to the polynucleotide of b), to form a chimeric polynucleotide sequence encoding a chimeric OspC-OspA protein such that OspC is upstream of OspA.
- 33. The method of claim 32, wherein the chimeric OspC-OspA protein has an amino acid sequence selected from the group consisting of: SEQ ID NOs: 146, 148, 150, 152, 154, 156, 158, 160, 162, 164, 166, 168, 170, 172, 174, 176, 178 and 180.
- 34. A method for producing a chimeric protein further comprising maintaining a host cell comprising the chimeric polynucleotide of claim 32 under conditions suitable for expression of the polynucleotide.
- 35. A method of delivering a chimeric protein comprising at least a first and a second polypeptide, wherein the first polypeptide comprises Borrelia burgdorferi OspC and wherein the second polypeptide comprises Borrelia burgdorferi OspA, such that OspC comprises the N-terminus of the protein, comprising administering the chimeric protein to an animal.
- 36. The method of claim 35, wherein the chimeric protein is administered with a physiologically-acceptable vehicle or carrier.
- 37. The method of claim 35, wherein the chimeric protein is encoded by a nucleic acid selected from the group consisting of: SEQ ID NOs: 145, 147, 149, 151, 153, 155, 157, 159, 161, 163, 165, 167, 169, 171, 173, 175, 177 and 179.
- 38. The method of claim 35, wherein the chimeric protein comprises an amino acid sequence selected from the group consisting of: SEQ ID NOs: 146, 148, 150, 152, 154, 156, 158, 160, 162, 164, 166, 168, 170, 172, 174, 176, 178 and 180.
- 39. A method of detecting a chimeric protein comprising at least a first and a second polypeptide, wherein the first polypeptide comprises Borrelia burgdorferi OspC and wherein the second polypeptide comprises Borrelia burgdorferi OspA, such that OspC comprises the N-terminus of the protein, comprising;
a) contacting a host sample with the chimeric protein, under conditions wherein antibodies, if present, in the host sample bind to the chimeric protein forming antigen antibody complexes; and b) detecting the antigen antibody complexes.
- 40. The method of claim 39, wherein the chimeric protein is encoded by a nucleic acid selected from the group consisting of: SEQ ID NOs: 145, 147, 149, 151, 153, 155, 157, 159, 161, 163, 165, 167, 169, 171, 173, 175, 177 and 179.
- 41. The method of claim 39, wherein the chimeric protein comprises an amino acid sequence selected from the group consisting of: SEQ ID NOs: 146, 148, 150, 152, 154, 156, 158, 160, 162, 164, 166, 168, 170, 172, 174, 176, 178 and 180.
- 42. A method of immunizing an animal or human against Lyme disease, comprising administering a composition comprising a chimeric protein of at least a first and a second polypeptide, wherein the first polypeptide comprises Borrelia burgdorferi OspC and wherein the second polypeptide comprises Borrelia burgdorferi OspA, such that OspC comprises the N-terminus of the protein.
- 43. The method of claim 42, wherein the chimeric protein comprises an amino acid sequence selected from the group consisting of: SEQ ID NOs: 146, 148, 150, 152, 154, 156, 158, 160, 162, 164, 166, 168, 170, 172, 174, 176, 178 and 180.
- 44. A method of immunizing an animal against Lyme disease, comprising administering a composition comprising a nucleic acid that encodes a chimeric protein comprising a first and a second polypeptide, wherein the first polypeptide comprises Borrelia burgdorferi OspC and wherein the second polypeptide comprises Borrelia burgdorferi OspA, such that OspC comprises the N-terminus of the protein.
- 45. The method of claim 44, wherein the nucleic acid encoding the chimeric protein is selected from the group consisting of: SEQ ID NOs: 145, 147, 149, 151, 153, 155, 157, 159, 161, 163, 165, 167, 169, 171, 173, 175, 177 and 179.
- 46. A physiological composition to vaccinate against and treat Borrelia infection in animals or humans, the composition comprising;
a) a chimeric protein comprising at least a first and a second polypeptide, wherein the first polypeptide comprises Borrelia burgdorferi OspC and wherein the second polypeptide comprises Borrelia burgdorferi OspA, such that OspC comprises the N-terminus of the protein, the amount being effective to ellicit an immune response to Borrelia burgdorferi; b) a physiologically-acceptable carrier or vehicle; and c) an adjuvant.
- 47. The composition of claim 46, wherein the chimeric protein is selected from the group consisting of: SEQ ID NOs: 146, 148, 150, 152, 154, 156, 158, 160, 162, 164, 166, 168, 170, 172, 174, 176, 178 and 180.
- 48. A physiological composition to vaccinate against and treat Borrelia infection in animals or humans, the composition comprising;
a) a nucleic acid that encodes a chimeric protein comprising at least a first and a second polypeptide, wherein the first polypeptide comprises Borrelia burgdorferi OspC and wherein the second polypeptide comprises Borrelia burgdorferi OspA, such that OspC comprises the N-terminus of the protein, the amount being effective to elicit an immune response to Borrelia burgdorferi; and b) a physiologically-acceptable carrier or vehicle.
- 49. The composition of claim 47, wherein the nucleic acid is selected from the group consisting of: SEQ ID NOs: 145, 147, 149, 151, 153, 155, 157, 159, 161, 163, 165, 167, 169, 171, 173, 175, 177 and 179.
- 50. A kit comprising;
a) a chimeric protein comprising at least a first and a second polypeptide, wherein the first polypeptide comprises Borrelia burgdorferi OspC and wherein the second polypeptide comprises Borrelia burgdorferi OspA, such that OspC comprises the N-terminus of the protein; and b) reagents for detecting antibody antigen complexes formed between the chimeric protein and antibodies, if present, in a user-supplied host sample.
- 51. The kit of claim 50, wherein the chimeric protein is selected from the group consisting of: SEQ ID NOs: 146, 148, 150, 152, 154, 156, 158, 160, 162, 164, 166, 168, 170, 172, 174, 176, 178 and 180.
