Claims
- 1. A compound having the structure: ##STR53## and the pharmaceutically-acceptable salts, esters and amides thereof, wherein:
- A is: hydrogen, ##STR54## Z is: sulfur, nitrogen or oxygen; X is: hydrogen, halogen, nitro, lower alkyl, lower alkyl substituted by halogen or nitro, aralkyl, alkaryl, or aralkyl or alkaryl substituted by hydrogen, halogen, nitro, lower alkyl or lower alkyl substituted by halogen or nitro;
- R, R.sup.1, R.sup.2, R.sup.3, R.sup.4, R.sup.6, R.sup.7, R.sup.9 and R.sup.10 are: independently hydrogen or alkyl, with the proviso that R.sup.6, R.sup.7, R.sup.9 and R.sup.10 are not each hydrogen;
- R.sup.5 is: amino, hydroxy, alkoxy, alkylamino, dialkylamino or alkoxyaryl; and
- R.sup.8 is: hydrogen, alkyl, alkyl carbonyl, alkoxy carbonyl, amino carbonyl, alkylaminocarbonyl or dialkylamino carbonyl, with any of the foregoing R.sup.8 substituents being aryl substituted.
- 2. A compound of claim 1 wherein A is ##STR55## and X is hydrogen or halogen.
- 3. A compound of claim 2 wherein R.sup.1, R.sup.2, R.sup.3 and R.sup.4 are each independently hydrogen or methyl.
- 4. A compound of claim 3 wherein R.sup.8 is hydrogen, lower alkyl, alkyl carbonyl, alkoxy carbonyl or alkylaminocarbonyl.
- 5. A compound of claim 4 wherein R.sup.8 is hydrogen, methyl, methyl carbonyl, lower alkoxy carbonyl or aminocarbonyl.
- 6. A compound of claim 5 wherein R.sup.6, R.sup.7, R.sup.9 and R.sup.10 are each independently hydrogen or lower alkyl.
- 7. A compound of claim 6 wherein R.sup.6, R.sup.7, R.sup.9 and R.sup.10 are each independently hydrogen or methyl.
- 8. A compound of claim 7 wherein R is hydrogen.
- 9. A compound of claim 8 wherein two of R.sup.6, R.sup.7, R.sup.9 and R.sup.11 are hydrogen and two of R.sup.6, R.sup.7, R.sup.9 and R.sup.10 are methyl.
- 10. A compound of claim 8 wherein one of R.sup.6, R.sup.7, R.sup.9 and R.sup.10 is hydrogen and the rest of R.sup.6, R.sup.7, R.sup.9 and R.sup.10 are each methyl.
- 11. A compound of claim 8 wherein one of R.sup.6, R.sup.7, R.sup.9 and R.sup.10 is methyl and the rest of R.sup.6, R.sup.7, R.sup.9 and R.sup.10 are each hydrogen.
- 12. A compound of claim 8 wherein A is ##STR56##
- 13. A compound of claim 12 wherein R.sup.8 is hydrogen, methyl, methoxy, --COOCH.sub.2 CH.sub.2 CH.sub.3 or --CONHCH.sub.3.
- 14. A compound of claim 13 wherein R.sup.5 is hydroxy, methoxy, --NH.sub.2 or --NHCH.sub.2 CH.sub.3.
- 15. A compound of claim 1 wherein the compound is:
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5)-disulfide;
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-L-valine, cyclic (2.fwdarw.5)-disulfide;
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-4-fluoro-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5)-disulfide;
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valinamide, cyclic (2.fwdarw.5)-disulfide;
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5)-disulfide, methyl ester;
- 2,3,6-trimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5) disulfide;

- 16. A compound of claim 15 wherein the compound is:
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5)-disulfide;
- 2. 6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valinamide, cyclic (2.fwdarw.5)-disulfide;
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5)-disulfide, methyl ester;
- 2,3,6-trimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5) disulfide;

- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5)-disulfide;
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valinamide, cyclic (2.fwdarw.5)-disulfide;
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5)-disulfide, methyl ester; or
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-4-fluoro-L-phenylalanyl-3-mercapto-D-valinamide, cyclic (2.fwdarw.5)-disulfide.
- 18. A compound of claim 1, having the structure: ##STR57##

- 25. A pharmaceutical composition comprising a pharmaceutically-acceptable carrier and a compound of claim 1.
- 26. The pharmaceutical composition of claim 25 wherein the compound is:
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5)-disulfide;
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valinamide, cyclic (2.fwdarw.5)-disulfide;
- 2. 6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5)-disulfide, methyl ester; or
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-4-fluoro-L-phenylalanyl-3-mercapto-D-valinamide, cyclic (2.fwdarw.5)-disulfide.
- 27. A method for treating pain in an animal comprising administering to said animal a therapeutically-effective amount of a compound of claim 1, said compound exhibiting activity systemically.
- 28. The method of claim 27 wherein the compound is:
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5)-disulfide;
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valinamide, cyclic (2.fwdarw.5)-disulfide;
- 2. 6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-L-phenylalanyl-3-mercapto-D-valine, cyclic (2.fwdarw.5)-disulfide, methyl ester; or
- 2,6-dimethyl-L-tyrosyl-3-mercapto-D-valylglycyl-4-fluoro-L-phenylalanyl-3-mercapto D valinamide, cyclic (2.fwdarw.5)-disulfide.
BACKGROUND OF THE INVENTION
1. Field of the Invention
This application is a continuation-in-part of application U.S. Ser. No. 497,833, filed on Mar. 22, 1990, entitled "Substituted Cyclic Penicillanic Acid Pentapeptides," now abandoned, which is a continuation-in-part of application U.S. Ser. No. 451,290, filed on Dec. 15, 1989, entitled "Substituted Cyclic Penicillanic Acid Pentapeptides," now abandoned.
