Claims
- 1. A method of maintaining separated viable tissue in proximate relationship, said method comprising:
applying to said separated viable tissue to hold together said tissue when said separated tissue is in contiguous relationship, a precursor composition comprising a polyfunctional crosslinking agent and a protein polymer comprising at least 40 weight % of repetitive units of from 3 to 15 amino acids of at least one naturally occurring structural protein, where there are at least two heteroatom functional groups present in said protein polymer, which heteroatom functional groups react with said crosslinking agent, whereby said precursor composition sets up to a strongly adherent adhesive composition; with said separated viable tissue held in proximate relationship.
- 2. A method according to claim 1, wherein said heteroatom functional group is amino.
- 3. A method according to claim 1, wherein said heteroatom functional group is as a result of reacting a naturally occurring amino acid in said protein polymer with a polyfunctional compound without setting up of said composition.
- 4. A method according to claim 1, wherein said units are selected from the group consisting of GAGAGS (SEQ ID NO:01), GVGVP (SEQ ID NO:02) and GXX, wherein the X's are the same or different and X is any amino acid, at least 10 number % and not more than 60 number % of X's being proline.
- 5. A method according to claim 4, wherein said units are GAGAGS (SEQ ID NO:01) and GVGVP (SEQ ID NO:02) and are present to form a block copolymer protein.
- 6. A method according to claim 1, wherein a difunctional compound capable of reacting with said polyfunctional crosslinking compound is included in said applying.
- 7. A method according to claim 6, wherein said difunctional compound is lysine or arginine.
- 8. A method of maintaining separated viable tissue in proximate relationship, said method comprising:
applying to said separated viable tissue to hold together said tissue when said separated tissue is in contiguous relationship, a precursor composition comprising a polyfunctional crosslinking agent reactive with amino groups and a protein block copolymer comprising at least 70 weight % of repetitive units of GAGAGS (SEQ ID NO:01) and GVGVP (SEQ ID NO:02), where in at least two units an amino acid is substituted with at least one of lysine or arginine or at least two intervening groups comprises at least one of lysine or arginine, whereby said precursor composition sets up to a strongly adherent adhesive composition; with said separated viable tissue held in proximate relationship.
- 9. A method according to claim 8, wherein the equivalent weight of said protein per lysine and arginine is in the range of 1 to 40 kD.
- 10. A method according to claim 8, wherein each block of said block copolymer has at least two units and the ratio of GAGAGS (SEQ ID NO:01) to GVGVP (SEQ ID NO:02) is in the range of 1:1-16.
- 11. A method according to claim 8, wherein said crosslinking agent has a plurality of amino reactive groups selected from the group consisting of aldehydo, isocyanate, thioisocyanate and activated carboxy.
- 12. A method according to claim 11, wherein said reactive group is isocyanate.
- 13. A method according to claim 8, wherein at least one of lysine or arginine is included in said applying.
- 14. A method of maintaining separated viable tissue in proximate relationship, said method comprising:
applying to said separated viable tissue to hold together said tissue when said separated tissue is in contiguous relationship, a precursor composition comprising a polyfunctional crosslinking agent selected from the group consisting of glutaraldehyde and polymethylene diisocyanate and a protein block copolymer of at least 30 kD comprising at least 70 weight % of repetitive units of GAGAGS (SEQ ID NO:01) and GVGVP (SEQ ID NO:02), where in at least two units an amino acid is substituted with at least one of lysine or arginine, said copolymer having a lysine and arginine equivalent weight in the range of 3 to 15 kD, whereby said precursor composition sets up to a strongly adherent adhesive composition; with said separated viable tissue held in proximate relationship.
- 15. A method of sealing a defect in viable tissue, said method comprising:
applying to said defect a precursor composition comprising a polyfunctional crosslinking agent and a protein polymer comprising at least 40 weight % of repetitive units of from 3 to 15 amino acids of at least one naturally occurring structural protein, where in at least two units an amino acid is substituted by a heteroatom functional group containing amino acid or at least two intervening groups comprise a heteroatom functional group, which heteroatom functional group reacts with said crosslinking agent, whereby said precursor composition sets up to a strongly adherent adhesive composition; whereby said defect is sealed.
- 16. A method according to claim 15, wherein said units are selected from the group consisting of GAGAGS (SEQ ID NO:01), GVGVP (SEQ ID NO:02) and GXX, wherein the X's are the same or different and X is any amino acid, at least 10 number % and not more than 60 number % of X's being proline.
- 17. A method according to claim 15, wherein said units are GAGAGS (SEQ ID NO:01) and GVGVP (SEQ ID NO:02) and are present to form a block copolymer protein.
- 18. A method according to claim 15, wherein said tissue is the wall of a vessel selected from an artery, vein, capillary, lung, dura or colon.
- 19. A kit comprising a polyfunctional crosslinking agent and a protein polymer comprising at least 40 weight % of repetitive units of from 3 to 15 amino acids of at least one naturally occurring structural protein, where in at least two units an amino acid is substituted by a heteroatom functional group containing amino acid, which heteroatom functional group reacts with said crosslinking agent.
- 20. A kit according to claim 19, wherein said heteroatom functional group is amino.
- 21. A kit according to claim 19, wherein said heteroatom functional group is as a result of reacting a naturally occurring amino acid in said protein polymer with a polyfunctional compound without setting up of said composition.
- 22. A kit according to claim 19, wherein said units are selected from the group consisting of GAGAGS (SEQ ID NO:01), GVGVP (SEQ ID NO:02) and GXX, wherein the X's are the same or different and X is any amino acid, at least 10 number % and not more than 60 number % of X's being proline.
- 23. A kit according to claim 22, wherein said units are GAGAGS (SEQ ID NO:01) and GVGVP (SEQ ID NO:02) and are present to form a block copolymer protein.
- 24. A kit according to claim 19, wherein said crosslinking agent has a plurality of reactive groups selected from the group consisting of aldehyde, isocyanate, thioisocyante and activated carboxy.
- 25. A kit comprising a protein block copolymer of at least 30 kD comprising at least 70 weight % of repetitive units of GAGAGS (SEQ ID NO:01) and GVGVP (SEQ ID NO:02), where in at least two units an amino acid is substituted with at least one of lysine or arginine, said copolymer having a lysine and arginine equivalent weight in the range of 1 to 40 kD and a crosslinking agent selected from the group consisting of glutaraldehyde and aliphatic diisocyanates.
- 26. A crosslinked protein composition, said protein prior to crosslinking being characterized by comprising at least 40 weight % of repetitive units of from 3 to 15 amino acids of at least one naturally occurring structural protein, where in at least two units an amino acid is substituted by a heteroatom functional group containing amino acid, said heteroatom functional group capable of reacting with at least one of aldehydo, isocyanate, thioisocyante and activated carboxy.
- 27. A crosslinked protein composition according to claim 26, wherein said protein is further characterized by comprising at least 70 weight % of repetitive units of GAGAGS (SEQ ID NO:01) and GVGVP (SEQ ID NO:02), where in at least two units an amino acid is substituted with at least one of lysine or arginine, said copolymer having a lysine and arginine equivalent weight in the range of 1 to 20 kD.
- 28. A method of maintaining separated viable tissue in proximate relationship, said method comprising:
applying to said separated viable tissue to hold together said tissue when said separated tissue is in contiguous relationship, a precursor composition comprising (1) a polyfunctional crosslinking agent comprising 2-aminoethyl glycinate, cholinyl lysinate or 1,3-propanediyl glycinate reacted with a difunctional compound which reacts with amino groups, and (2) a protein polymer comprising at least 40 weight % of repetitive units of from 3 to 15 amino acids of at least one naturally occurring structural protein, where there are at least two amino functional groups present in said protein polymer, which amino functional groups react with said crosslinking agent, whereby said precursor composition sets up to a strongly adherent adhesive composition; with said separated viable tissue held in proximate relationship.
