Claims
- 1. An isolated DNA sequence encoding a mammalian TNF receptor (TNF-R).
- 2. An isolated DNA sequence according to claim I which encodes a human TNF-R.
- 3. An isolated DNA sequence according to claim 1, selected from the group consisting of:
(a) cDNA clones having a nucleotide sequence derived from the coding region of a native mammalian TNF-R gene; (b) DNA sequences capable of hybridization to the clones of (a) under moderately stringent conditions and which encode biologically active TNF-R protein; and (c) DNA sequences which are degenerate as a result of the genetic code to the DNA sequences defined in (a) and (b) and which encode biologically a TNF-R protein.
- 4. An isolated DNA sequence according to claim 2 which encodes a soluble human TNF-R.
- 5. An isolated DNA sequence according to claim 4 which encodes a soluble human TNF-R comprising the sequence of amino acids 1 through x of FIG. 2A, wherein x is selected from the group consisting of amino acids 163-235.
- 6. An isolated DNA sequence according to claim 5 which encodes a soluble human TNF-R comprising the sequence of amino acids 1-235 of FIG. 2A.
- 7. An isolated DNA sequence according to claim 5 which encodes a soluble human TNF-R comprising the sequence of amino acids 1-185 of FIG. 2A.
- 8. An isolated DNA sequence according to claim 5 which encodes a soluble human TNF-R comprising the sequence of amino acids 1-163 of FIG. 2A
- 9. An isolated DNA sequence according to claim 1 encoding the TNF-R polypeptide expressed by pCAV/NOT-TNF-R (ATCC 68088).
- 10. A recombinant expression vector comprising a DNA sequence according to claim 1.
- 11. A recombinant expression vector comprising a DNA sequence according to claim 2.
- 12. A recombinant expression vector comprising a DNA sequence according to claim 3.
- 13. A recombinant expression vector comprising a DNA sequence according to claim 4.
- 14. A recombinant expression vector comprising a DNA sequence according to claim 5.
- 15. A recombinant expression vector comprising a DNA sequence according to claim 6.
- 16. A recombinant expression vector comprising a DNA sequence according to claim 7.
- 17. A recombinant expression vector comprising a DNA sequence according to claim 8.
- 18. A recombinant expression vector comprising a DNA sequence according to claim 9.
- 19. A process for preparing a mammalian TNF receptor or an analog thereof, comprising culturing a suitable host cell comprising a vector according to claim 10 under conditions promoting expression.
- 20. A process for preparing a human TNF receptor or an analog thereof, comprising culturing a suitable host cell comprising a vector according to claim 11 under conditions promoting expression.
- 21. A process for preparing a human TNF receptor or an analog thereof, comprising culturing a suitable host cell comprising a vector according to claim 12 under conditions promoting expression.
- 22. A process for preparing a human TNF receptor or an analog thereof, comprising culturing a suitable host cell comprising a vector according to claim 13 under conditions promoting expression.
- 23. A process for preparing a human TNF receptor or an analog thereof, comprising culturing a suitable host cell comprising a vector according to claim 14 under conditions promoting expression.
- 24. A process for preparing a human TNF receptor or an analog thereof, comprising culturing a suitable host cell comprising a vector according to claim 15 under conditions promoting expression.
- 25. A process for preparing a human TNF receptor or an analog thereof, comprising culturing a suitable host cell comprising a vector according to claim 16 under conditions promoting expression.
- 26. A process for preparing a human TNF receptor or an analog thereof, comprising culturing a suitable host cell comprising a vector according to claim 17 under conditions promoting expression.
- 27. A process for preparing a human TNF receptor or an analog thereof, comprising culturing a suitable host cell comprising a vector according to claim 18 under conditions promoting expression.
- 28. An isolated and purified biologically active mammalian TNF receptor composition.
- 29. An isolated and purified TNF receptor composition according to claim 28, consisting essentially of human TNF receptor.
- 30. An isolated and purified TNF receptor composition according to claim 28, consisting essentially of soluble human TNF receptor.
- 31. An isolated and purified TNF receptor composition according to claim 30, comprising the sequence of amino acids 1-235 of FIG. 2A.
- 32. An isolated and purified TNF receptor composition according to claim 30, comprising the sequence of amino acids 1-185 of FIG. 2A.
- 33. An isolated and purified TNF receptor composition according to claim 30, comprising the sequence of amino acids 1-163 of FIG. 2A.
- 34. A composition for regulating immune or inflammatory responses in a mammal, comprising an effective amount of a mammalian TNF receptor protein composition according to claim 28, and a suitable diluent or carrier.
- 35. A method for regulating immune responses in a mammal, comprising administering an effective amount of a composition according to claim 34.
- 36. An assay method for detection of TNF or TNF receptor molecules or the interaction thereof, comprising use of a protein composition according to claim 28.
- 37. Antibodies immunoreactive with mammalian TNF receptors.
