Claims
- 1. A method for the protection of a poikolothermic fish against infection by the bacterial pathogen Piscirickettsia salmonis comprised of administering intracellularly to said animal an immunogenic amount of a pharmaceutical composition comprising at least one of a principal antigen, an OspA lipoprotein, its variants, its non-lipidated form, or antigenic peptides derived or synthesized thereof.
- 2. The method of claim 1 wherein the principal antigen, the OspA lipoprotein, its variants, its non-lipidated form, or antigenic peptides derived or synthesized thereof are fused to at least one other protein, protein fragment or peptide either at the N or C terminus or both where that protein, protein fragment or peptide may be used to facilitate expression and the formation of one of insoluble intracellular aggregates (inclusion bodies), soluble intracellular protein, soluble extracellular protein, or soluble periplasmic protein or to facilitate either expression or the formation of one of insoluble intracellular aggregates (inclusion bodies), soluble intracellular protein, soluble extracellular protein, or soluble periplasmic protein.
- 3. The method of claim 2, wherein said pharmaceutical composition is encapsulated in or adsorbed on or absorbed in or are in the form of an insoluble polymeric matrix.
- 4. The method of claim 2, wherein the principle OspA antigen, its variants, fragments, fusion proteins, or synthetic peptides thereof comprise sequence homologues of the OspA protein.
- 5. The method of claim 4 where the pharmaceutical composition is formulated with an adjuvant.
- 6. The method of claim 2 where the route of administration is either orally, intraperitoneally, intramuscularly, intradermally or by immersion or spraying or by any combination of these methods.
- 7. The method of claim 2, where the principle antigen is encoded by OspA or an OspA homologue where the sequence has been optimized for expression in a suitable expression host microorganism such as an E. coli bacterial strain.
- 8. The method of claim 2 where the expression of the OspA protein, its variants, lipoprotein version or antigenic peptides derived or synthesized thereof in E. coli is effected by a promoter.
- 9. A method for the protection of a poikolothermic fish against infection by the bacterial pathogen Piscirickettsia salmonis comprised of administering intracellularly to said animal an immunogenic amount of a pharmaceutical composition comprising at least one of a DNA sequence corresponding to that of ospA, fragments or synthetic oligonucleotides thereof or of DNA sequence homologues of ospA, or fragments or synthetic oligonucleotides derived thereof.
- 10. The method of claim 9 wherein the DNA sequence corresponding to that of ospA, fragments or synthetic oligonucleotides thereof or of DNA sequence homologues of ospA, or fragments or synthetic oligonucleotides derived thereof is fused to at least one other DNA fragment, where the DNA fragment is used to facilitate expression.
- 11. The method of claim 9, wherein said pharmaceutical composition are encapsulated in or absorbed in or adsorbed to or are in the form of an insoluble polymeric matrix.
- 12. The method of claim 9 where the pharmaceutical composition is formulated with an adjuvant.
- 13. The method of claim 9 where the route of administration is either orally, intraperitoneally, intramuscularly, intradermally or by immersion or spraying or by any combination of these methods.
