Claims
- 1. A composition, comprising:
- a water-insoluble biocompatible material comprising a modified hyaluronic acid in which at least about 5% of carboxyl functionalities have been derivatized, and wherein essentially all of the derivatized functionalities are O-acyl or N-acyl ureas; and
- a substance having pharmaceutical activity covalently bonded to said modified hyaluronic acid and releasable therefrom by hydrolysis.
- 2. The composition of claim 1, wherein said modified hyaluronic acid is formed by reacting hyaluronic acid, or a salt thereof, with a monocarbodiimide of the formula:
- R.sup.1 --N.dbd.C.dbd.N--R.sup.2
- in which R.sup.1 and R.sup.2 are each independently selected from the group consisting of hydrocarbyl, substituted-hydrocarbyl, alkoxy, aryloxy, and alkaryloxy.
- 3. The composition of claim 2, wherein said monocarbodiimide is selected from the group consisting of 3-(dimethylamino) propyl-3-ethylcarbodiimide, cyclohexyl-B-(N-methylomorpholino) ethylcarbodiimide p-toluene-sulfonate, and 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide methiodide.
- 4. The composition of claim 1, wherein the modified hyaluronic acid is formed by reacting hyaluronic acid, or a salt thereof, with a biscarbodiimide of the formula:
- R.sup.1 --N.dbd.C.dbd.N--R.sup.2 --N.dbd.C.dbd.N--R.sup.3
- in which R.sup.1, R.sup.2, and R.sup.3 are each independently selected from the group consisting of hydrocarbyl, substituted-hydrocarbyl, alkoxy, aryloxy, and alkaryloxy.
- 5. The composition of claim 4, wherein said biscarbodiimide is one of phenylenebis-(ethyl)-carbodiimide and 1,6-hexamethylenebis (ethylcarbodiimide).
- 6. The composition of claim 2, wherein the molar equivalent ratio of said monocarbodiimide to said hyaluronic acid during said reaction is at least about 1:20.
- 7. The composition of claim 1, wherein said pharmaceutically-active substance is a drug.
- 8. The composition of claim 1, wherein said composition is a gel.
- 9. The composition of claim 1, wherein said composition is a film.
- 10. The composition of claim 1, wherein said composition is a sponge.
- 11. A composition, comprising:
- a water-insoluble biocompatible material comprising a modified hyaluronic acid in which at least about 5% of carboxyl functionalities have been derivatized, and wherein essentially all of the derivatized functionalities are O-acyl or N-acyl ureas; and
- a substance having pharmaceutical activity dispersed within said material.
- 12. The composition of claim 11, wherein said modified hyaluronic acid is formed by reacting hyaluronic acid, or a salt thereof, with a monocarbodiimide of the formula:
- R.sup.1 --N.dbd.C.dbd.N--R.sup.2
- in which R.sup.1 and R.sup.2 are each independently selected from the group consisting of hydrocarbyl, substituted-hydrocarbyl, alkoxy, aryloxy, and alkaryloxy.
- 13. The composition of claim 12, wherein said monocarbodiimide is selected from the group consisting of 3-(Dimethylamino) propyl-3-ethylcarbodiimide, cyclohexyl-B-(N-methylomorpholino) ethylcarbodiimide p-toluene-sulfonate, and 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide methiodide.
- 14. The composition of claim 11, wherein the modified hyaluronic acid is formed by reacting hyaluronic acid or a salt thereof, with a biscarbodiimide of the formula:
- R.sup.1 --N.dbd.C.dbd.N--R.sup.2 --N.dbd.C.dbd.N--R.sup.3
- in which R.sup.1, R.sup.2, and R.sup.3 are each independently selected from the group consisting of hydrocarbyl, substituted-hydrocarbyl, alkoxy, aryloxy, and alkaryloxy.
- 15. The composition of claim 14, wherein said biscarbodiimide is one of phenylenebis-(ethyl)-carbodiimide and 1,6-hexamethylenebis (ethylcarbodiimide).
- 16. The composition of claim 12, wherein the molar equivalent ratio of said monocarbodiimide to said hyaluronic acid is at least about 1:20.
- 17. The composition of claim 11, wherein said pharmaceutically-active substance is a drug.
- 18. The composition of claim 11, wherein said composition is a gel.
- 19. The composition of claim 11, wherein said composition is a film.
- 20. The composition of claim 11, wherein said composition is a sponge.
