Claims
- 1. A genetic vector capable of replication in the cytoplasm of a plant cell, the vector comprising a first subgenomic promoter operably linked to a first polynucleotide encoding a target gene inhibitory RNA, and a second subgenomic promoter operably linked to a second polynucleotide encoding a viral coat protein, wherein said first polynucleotide is upstream to said second polynucleotide, and said vector is capable of systemically infecting plants and producing the inhibitory RNA.
- 2. The vector according to claim 1, wherein the vector is derived from a plant RNA virus.
- 3. The vector according to claim 2, wherein the vector is derived from a single stranded RNA plant virus.
- 4. The vector according to claim 1, wherein at least one of the subgenomic promoters is derived from a tobamovirus.
- 5. The vector according to claim 4, wherein the first polynucleotide encodes a phytoene desaturase RNA in an antisense direction in relation to the first subgenomic promoter.
- 6. The vector according to claim 4, wherein the first polynucleotide encodes a phytoene synthase RNA in an antisense direction in relation to the first subgenomic promoter.
- 7. A method of producing a plant cell having reduced expression of a gene of interest, the method comprising the steps of transfecting a pant cell with a genetic vector according to claim 1, wherein the target gene inhibitory, RNA is specific for the gene of interest.
- 8. The method according to claim 7, wherein the vector is derived form a plant RNA virus.
- 9. The method according to claim 8, wherein the vector is derived form a single stranded RNA plant virus.
- 10. The method according to claim 9, wherein at least one of the subgenomic promoters is derived from a tobamovirus.
- 11. A method of producing a plant cell having reduced expression of a gene of interest, the method comprising the steps of transfecting a cell with the genetic vector according to claim 5, and then growing the transfected cell under conditions suitable for growth of the vector.
- 12. A method of producing a plant cell having reduced expression of a gene of interest, the method comprising the steps of transfecting a cell with the genetic vector according to claim 6, and then growing the transfected cell under conditions suitable for growth of the vector.
- 13. A plant cell produced by the method of claim 7.
- 14. The plant cell according to 13, wherein the vector is derived from a plant RNA virus.
- 15. The plant cell according to claim 14, wherein the vector is derived from a single stranded RNA plant virus.
- 16. The plant cell according to claim 15, wherein at least one of the subgenomic promoters is derived from a tobamovirus.
- 17. A plant cell comprising the vector according to claim 1.
- 18. The plant cell according to claim 17, wherein the vector is derived from a plant RNA virus.
- 19. The plant cell according to claim 18, wherein the vector is derived from a single stranded RNA plant virus.
- 20. The plant cell according to claim 19, wherein at least one of the subgenomic promoters is derived from a tobamovirus.
- 21. A plant cell comprising the vector according to claim 5.
- 22. The vector according to claim 1, wherein said target gene inhibitory RNA is an endogenous plant gene inhibitory RNA.
- 23. The vector according to claim 1, wherein said target gene inhibitory RNA is an anti-sense RNA.
- 24. The vector according to claim 1, wherein said target gene inhibitory RNA is a co-suppressor RNA.
