Claims
- 1. A nucleic acid molecule encoding the heavy chain variable region of an antibody having specificity for human interleukin-5, wherein said heavy chain variable region comprises in sequential order: 1) complementarity determining region (CDR) 1, comprising the amino acid sequence identified as SEQ ID NO: 7; 2) CDR 2 comprising the amino acid sequence identified as SEQ ID NO: 8, and; 3) CDR 3 comprising the amino acid sequence selected from the group consisting of:
- (A) SEQ ID NO: 9; and
- (B) SEQ ID NO: 14.
- 2. A nucleic acid molecule encoding the light chain variable region of an antibody having specificity for human interleukin-5, wherein said light chain variable region comprises in sequential order: 1 ) CDR 1, comprising the amino acid sequence identified as SEQ ID NO: 10; 2) CDR 2, comprising the amino acid sequence identified as SEQ ID NO: 11, and; 3) CDR 3 comprising the amino acid sequence selected from the group consisting of:
- (A) SEQ ID NO: 12;
- (B) SEQ ID NO: 13;
- (C) SEQ ID NO: 47; and
- (D) SEQ ID NO: 48.
- 3. An isolated nucleic acid sequence which is selected from the group consisting of:
- (A) a nucleic acid sequence encoding a synthetic antibody having specificity for human interleukin-5, wherein said antibody comprises a heavy chain and a light chain, and the framework regions of said heavy and light chains are obtained from at least a first antibody and the amino acid sequences of the complementarity determining regions of said heavy chain are comprised in sequential order: 1) CDR 1, comprising the amino acid sequence identified as SEQ ID NO: 7; 2) CDR 2, comprising the amino acid sequence identified as SEQ ID NO: 8, and; 3) CDR 3 comprising the amino acid sequence selected from the group consisting of:
- (i) SEQ ID NO: 9; and
- (ii) SEQ ID NO: 14, and
- (B) a fragment of the nucleic acid sequence of (A) which encodes a protein having a dissociation constant equal or less than about 3.5.times.10.sup.11 M for human interleukin-5.
- 4. The isolated nucleic acid sequence according to claim 3 wherein the sequence encoding the humanized heavy chain variable region comprising the nucleic acid sequence of SEQ ID NO: 18.
- 5. The isolated nucleic acid sequence according to claim 3 wherein the sequence encoding the humanized heavy chain variable region comprising the nucleic acid sequence of SEQ ID NO: 61.
- 6. The isolated nucleic acid sequence according to claim 3 wherein the sequence encoding the humanized light chain variable region comprising the nucleic acid sequence of SEQ ID NO: 20.
- 7. The isolated nucleic acid sequence according to claim 3 wherein the sequence encoding the humanized light chain variable region comprising the nucleic acid sequence of SEQ ID NO: 69.
- 8. A recombinant plasmid comprising the isolated nucleic acid sequence of claim 3.
- 9. A host cell transfected with the recombinant plasmid of claim 8.
- 10. A process for producing a synthetic antibody specific for human interleukin-5 comprising culturing a cell line transfected with the recombinant plasmid of claim 8 under the control of selected regulatory sequences capable of directing the expression thereof in said cell line.
- 11. The nucleic acid molecule according to claim 3, wherein the amino acid sequences of the complementarity determining regions of the light chain of said synthetic antibody are:
- iii. SEQ ID NO: 10, 11, 12; or
- iv. SEQ ID NO: 10, 11, 13.
- 12. An isolated nucleic acid sequence which is selected from the group consisting of:
- (a) a nucleic acid sequence encoding the a synthetic antibody comprising a heavy chain and a light chain, wherein the framework regions of said heavy and light chains are obtained from at least a first antibody and the amino acid sequences of the complementarity determining regions of the heavy chain are obtained from a monoclonal antibody selected from the group consisting of monoclonal antibody 24G9, produced by the hybridoma ATCC 11780; monoclonal antibody, 4A6, produced by the hybridoma ATCC HB11943; and monoclonal antibody, 5D3, produced by the hybridoma ATCC HB11942;
- (b) a fragment of (a) which encodes a protein having a dissociation constant equal or less than about 3.5.times.10.sup.-11 M for human interleukin-5.
- 13. A recombinant plasmid comprising the isolated nucleic acid sequence of claim 12.
- 14. A host cell transfected with the recombinant plasmid of claim 13.
- 15. A process for producing a synthetic antibody specific for human interleukin-5 comprising culturing a cell line transfected with the recombinant plasmid of claim 13 under the control of selected regulatory sequences capable of directing the expression thereof in said cell line.
