Claims
- 1. A method for selective ex-vivo expansion of stem cells, comprising the steps of;
- (a) culturing stem cells in a growth media, comprising;
- a human interleukin-3 mutant polypeptide, having at least three times greater cell proliferative activity than native human interleukin-3, in at least one assay selected from the group consisting of: AML cell proliferation, TF-1 cell proliferation and Methylcellulose assay; said polypeptide comprising a sequence selected from the group consisting of:
- (i) the sequence of SEQ ID NO:1; and
- (ii) an N-terminal methionine residue, alanine residue or methionine-alanine di-peptide immediately preceding said sequence according to (i);

- (b) harvesting said cultured cells.

The method according to claim 6, wherein said factor is selected from the group consisting of GM-CSF, G-CSF, Meg-CSF, M-CSF, erythropoietin (EPO), IL-1, IL-4, IL-2, IL-5, IL-6, IL-7, IL-8, IL-9, IL-10, IL-11, IL-12, IL-13, LIF, flt3/ligand, human growth hormone, B-cell growth factor, B-cell differentiation factor, eosinophil differentiation factor and stem cell factor (SCF).
- 8. The method according to claim 7, wherein said factor is G-CSF or GM-CSF.
- 9. The method according to claim 1, 2, 3, 4, or 5, wherein said factor is selected from the group consisting of GM-CSF, G-CSF, Meg-CSF, M-CSF, erythropoietin (EPO), IL-1, IL-4, IL-2, IL-5, IL-6, IL-7, IL-8, IL-9, I:L-10, IL-11, IL-12, IL-13, LIF, flt3/ligand, human growth hormone, B-cell growth factor, B-cell differentiation factor, eosinophil differentiation factor and stem cell factor (SCF).
- 10. The method according to claim 9, wherein said factor is G-CSF or GM-CSF.
- 11. The method according to claim 6 wherein said modified human interleukin-3 polypeptide has at least five times greater cell proliferative activity than native human interleukin-3.
- 12. The method according to claim 6 wherein said modified human interleukin-3 polypeptide has at least ten times greater cell proliferative activity than native human interleukin-3.
- 13. The method according to claim 7 wherein said modified human interleukin-3 polypeptide has at least five times greater cell proliferative activity than native human interleukin-3.
- 14. The method according to claim 7 wherein said modified human interleukin-3 polypeptide has at least ten times greater cell proliferative activity than native human interleukin-3.
- 15. The method according to claim 9 wherein said modified human interleukin-3 polypeptide has at least five times greater cell proliferative activity than native human interleukin-3.
- 16. The method according to claim 9 wherein said modified human interleukin-3 polypeptide has at least ten times greater cell proliferative activity than native human interleukin-3.
- 17. The method according to claim 10 wherein said modified human interleukin-3 polypeptide has at least five times greater cell proliferative activity than native human interleukin-3.
- 18. The method according to claim 10 wherein said modified human interleukin-3 polypeptide has at least ten times greater cell proliferative activity than native human interleukin-3.
- 19. A method for the treatment of a patient having a hematopoietic disorder, comprising the steps of;
- (a) removing stem cells from said patient;
- (b) culturing said stem cells in a growth media comprising;
- a human interleukin-3 mutant polypeptide, having at least three times greater cell proliferative activity than native human interleukin-3, in at least one assay selected from the group consisting of: AML cell proliferation, TF-1 cell proliferation and Methylcellulose assay; said polypeptide comprising a sequence selected from the group consisting of:

- 22. A method for the treatment of a patient having a hematopoietic disorder, comprising the steps of;
- (a) removing stem cells from said patient;
- (b) culturing said stem cells in a growth media comprising;
- a human interleukin-3 mutant polypeptide, having at least three times greater cell proliferative activity than native human interleukin-3, in at least one assay selected from the group consisting of: AML cell proliferation, TF-1 cell proliferation and Methylcellulose assay; said polypeptide comprising a sequence selected from the group consisting of:
- (i) the sequence of SEQ ID NO:7; and
- (ii) an N-terminal methionine residue, alanine residue or methionine-alanine di-peptide immediately preceding said sequence according to (i);

- a factor selected from the group consisting of: a colony stimulating factor, a cytokine, a lymphokine, an interleukin, and a hematopoietic growth factor;
- (c) harvesting said cultured cells; and
- (d) transplanting said cultured cells into said patient.
- 23. A method for the treatment of a patient having a hematopoietic disorder, comprising the steps of;
- (a) removing stem cells from said patient;
- (b) culturing said stem cells in a growth media comprising;
- a human interleukin-3 mutant polypeptide, having at least three times greater cell proliferative activity than native human interleukin-3, in at least one assay selected from the group consisting of: AML cell proliferation, TF-1 cell proliferation and Methylcellulose assay; said polypeptide comprising a sequence selected from the group consisting of:
- (i) the sequence of SEQ ID NO:8; and
- (ii) an N-terminal methionine residue, alanine residue or methionine-alanine di-peptide immediately preceding said sequence according to (i);

- 24. The method according to claim 19, 20, 21, 22, or 23, further comprising the step of separating said stem cells from other cells before culturing said stem cells.
- 25. The method according to claim 24, wherein said factor is selected from the group consisting of GM-CSF, G-CSF, Meg-CSF, M-CSF, erythropoietin (EPO), IL-1, IL-4, IL-2, IL-5, IL-6, IL-7, IL-8, IL-9, IL-10, IL-ll, IL-12, IL-13, LIF, flt3/ligand, human growth hormone, B-cell growth factor, B-cell differentiation factor, eosinophil differentiation factor and stem cell factor (SCF).
- 26. The method according to claim 25, wherein said factor is G-CSF or GM-CSF.
- 27. The method according to claim 19, 20, 21, 22, or 23, wherein said factor is selected from the group consisting of GM-CSF, G-CSF, Meg-CSF, M-CSF, erythropoietin (EPO), IL-1, IL-4, IL-2, IL-5, IL-6, IL-7, IL-8, IL-9, IL-10, IL-11, IL-12, IL-13, LIF, flt3/ligand, human growth hormone, B-cell growth factor, B-cell differentiation factor, eosinophil differentiation factor and stem cell factor (SCF).
- 28. The method according to claim 27, wherein said factor is G-CSF or GM-CSF.
- 29. The method according to claim 24 wherein said modified human interleukin-3 polypeptide has at least five times greater cell proliferative activity than native human interleukin-3.
- 30. The method according to claim 24 wherein said modified human interleukin-3 polypeptide has at least ten times greater cell proliferative activity than native human interleukin-3.
- 31. The method according to claim 25 wherein said modified human interleukin-3 polypeptide has at least five times greater cell proliferative activity than native human interleukin-3.
- 32. The method according to claim 25 wherein said modified human interleukin-3 polypeptide has at least ten times greater cell proliferative activity than native human interleukin-3.
- 33. The method according to claim 26 wherein said modified human interleukin-3 polypeptide has at least five times greater cell proliferative activity than native human interleukin-3.
- 34. The method according to claim 26 wherein said modified human interleukin-3 polypeptide has at least ten times greater cell proliferative activity than native human interleukin-3.
