Claims
- 1. A method for protecting a recombinant AAV virion from loss of activity resulting from exposure of the virion to a cycle of freezing and thawing, said method comprising admixing the virion with a virion-stabilizing composition comprising a dihydric or polyhydric alcohol selected from the group consisting of polyethylene glycol wherein the polyethylene glycol is present at a concentration of 2 wt. % to 40 wt. %, propylene glycol wherein the propylene glycol is present at a concentration of 2 wt. % to 60 wt. %, and sorbitol wherein the sorbitol is present at a concentration of from 0.1 wt. % to 10 wt. %, whereby the recombinant AAV virion is protected from loss of activity from exposure to a cycle of freezing and thawing.
- 2. The method of claim 1, wherein the one or more alcohols is sorbitol.
- 3. The method of claim 1, wherein the one or more alcohols is polyethylene glycol.
- 4. The method of claim 1, wherein the one or more alcohols is propylene glycol.
- 5. The method of claim 1, wherein the virion-stabilizing composition further comprises a detergent.
- 6. The method of claim 5, wherein the detergent is a sorbitan ester.
- 7. The method of claim 6, wherein the sorbitan ester is selected from the group consisting of polyoxyethylenesorbitan monolaurate (TWEEN-20), polyoxyethylenesorbitan monopalmitate (TWEEN-40), polyoxyethylenesorbitan monostearate (TWEEN-60), polyoxyethylenesorbitan tristearate (TWEEN-65), polyoxyethylenesorbitan monooleate (TWEEN-80) and polyoxyethylenesorbitan trioleate (TWEEN-85).
- 8. The method of claim 1, wherein the recombinant AAV virion is provided as a lyophilized preparation.
- 9. A method for protecting a recombinant AAV virion from loss of activity resulting from exposure of the virion to a cycle of freezing and thawing, said method comprising admixing the virion with a virion-stabilizing composition comprising sorbitol and a sorbitan ester selected from the group consisting of polyoxyethylenesorbitan monolaurate (TWEEN-20) and polyoxyethylenesorbitan monooleate (TWEEN-80).
- 10. A method for protecting a recombinant AAV virion from loss of activity resulting from storage of the virion in a glass vessel comprising admixing the virion with a virion-stabilizing composition comprising a dihydric or polyhydric alcohol selected from the group consisting of polyethylene glycol wherein the polyethylene glycol is present at a concentration of 2 wt. % to 40 wt. %, propylene glycol wherein the propylene glycol is present at a concentration of 2 wt. % to 60 wt. %, and sorbitol wherein the sorbitol is present at a concentration of from 0.1 wt. % to 10 wt. %, whereby the recombinant AAV virion is protected from loss of activity from storage of the virion in a glass vessel.
- 11. The method of claim 10, wherein the one or more alcohols is sorbitol.
- 12. The method of claim 10, wherein the one or more alcohols is polyethylene glycol.
- 13. The method of claim 10, wherein the one or more alcohols is propylene glycol.
- 14. The method of claim 10, wherein the virion-stabilizing composition further comprises a detergent.
- 15. The method of claim 14, wherein the detergent is a sorbitan ester.
- 16. The method of claim 15, wherein the sorbitan ester is selected from the group consisting of polyoxyethylenesorbitan monolaurate (TWEEN-20), polyoxyethylenesorbitan monopalmitate (TWEEN-40), polyoxyethylenesorbitan monostearate (TWEEN-60), polyoxyethylenesorbitan tristearate (TWEEN-65), polyoxyethylenesorbitan monooleate (TWEEN-80) and polyoxyethylenesorbitan trioleate (TWEEN-85).
- 17. The method of claim 10, wherein the recombinant AAV virion is provided as a lyophilized preparation.
- 18. A method for protecting a recombinant AAV virion from loss of activity resulting from storage of the virion in a glass vessel comprising admixing the virion with a virion-stabilizing composition comprising sorbitol and a sorbitan ester selected from the group consisting of polyoxyethylenesorbitan monolaurate (TWEEN-20) and polyoxyethylenesorbitan monooleate (TWEEN-80).
- 19. The method of claim 1, wherein sorbitol is present at a concentration of about 1 wt. % to 5 wt. %.
- 20. The method of claim 1, wherein the polyethylene glycol is present at a concentration of about 10 wt. % to 25 wt. %.
- 21. The method of claim 1, wherein the propylene glycol is present at a concentration of 5 wt. % to 30 wt. %.
- 22. The method of claim 10, wherein sorbitol is present at a concentration of about 1 wt. % to 5 wt. %.
- 23. The method of claim 10, wherein the polyethylene glycol is present at a concentration of about 10 wt. % to 25 wt. %.
- 24. The method of claim 10, wherein the propylene glycol is present at a concentration of 5 wt. % to 30 wt. %.
