Claims
- 1. An isolated nucleic acid molecule comprising a nucleic acid sequence coding for a trkC-ligand-binding polypeptide comprising an amino acid sequence selected from the group consisting of:
- (a) mature human trkC having the sequence in SEQ ID NO:6;
- (b) an amino acid sequence encoded by a naturally-occurring splice variant of (a);
- (c) an amino acid sequence comprising the amino acid sequence 32 to 429 of SEQ ID NO:6;
- (d) an amino acid sequence comprising an extracellular domain of SEQ ID NO:6; and
- (e) a second immunoglobulin-like domain of (a), (b), (c), or (d).
- 2. An expression vector comprising the nucleic acid molecule of claim 1 operably linked to control sequences recognized by a host cell transformed with the vector.
- 3. A host cell transformed with the vector of claim 2.
- 4. A method of using a nucleic acid molecule according to claim 1 comprising expressing said nucleic acid molecule in a cultured host cell transformed with a vector comprising said nucleic acid molecule operably linked to control sequences recognized by the host cell, and recovering the encoded polypeptide from the host cell.
- 5. A method for producing a polypeptide according to claim 1 comprising inserting onto DNA of a cell containing nucleic acid encoding said polypeptide a transcription regulatory element in suffcient proximity and orientation to the nucleic acid molecule to influence the transcription thereof, and inducibly or constitutively expressing the nucleic acid encoding said polypeptide using the transcription regulatory element.
- 6. The nucleic acid of claim 1, wherein the polypeptide has a tyrosine kinase domain.
- 7. The nucleic acid of claim 1, wherein the polypeptide is devoid of a tyrosine kinase domain.
- 8. The nucleic acid of claim 1, wherein the polypeptide has a functional transmembrane domain.
- 9. The nucleic acid of claim 1, wherein the polypeptide is devoid of a functional transmembrane domain.
- 10. The nucleic acid molecule of claim 1, wherein the selected amino acid sequence is (a).
- 11. The nucleic acid of claim 1, wherein the polypeptide comprises amino acids 32 to 839 of the amino acid sequence set forth in SEQ. ID. NO:6.
- 12. The nucleic acid of claim 1, wherein the polypeptide comprises SEQ ID NO:8.
- 13. The nucleic acid molecule of claim 1, wherein the selected amino acid sequence is (b).
- 14. The nucleic acid molecule of claim 1, wherein the selected amino acid sequence is (c).
- 15. The nucleic acid molecule of claim 1, wherein the selected amino acid sequence is (d).
- 16. The nucleic acid molecule of claim 1, wherein the selected amino acid sequence is (e).
- 17. The nucleic acid of claim 11, wherein the polypeptide is devoid of amino acids ESTDNFILF (SEQ ID NO:36) at amino acid positions 402 to 410.
- 18. The nucleic acid of claim 11, wherein the polypeptide is devoid of amino acids LFNPSGNDFCIWCE (SEQ ID NO:37) at amino acid positions 712 to 725.
- 19. The nucleic acid of claim 11, wherein the polypeptide is unglycosylated.
- 20. The nucleic acid of claim 12, wherein the polypeptide is devoid of amino acids ESTDNFILF (SEQ ID NO:36) at amino acid positions 402 to 410.
- 21. The nucleic acid of any one of claims 11, 12, 17, 18, or 20, wherein the polypeptide is devoid of amino acids 430 to 453.
- 22. An isolated nucleic acid molecule selected from the group consisting of a nucleic acid sequence consisting of SEQ ID NO:1 and a nucleic acid sequence coding for a trkB-ligand-binding polypeptide comprising an amino acid sequence encoded by a naturally-occurring splice variant of SEQ ID NO:1.
- 23. An expression vector comprising the nucleic acid molecule of claim 22.
- 24. A host cell transformed with the vector of claim 23.
- 25. The nucleic acid of claim 22, wherein the polypeptide has a tyrosine kinase domain.
- 26. The nucleic acid of claim 22, wherein the polypeptide is devoid of a tyrosine kinase domain.
- 27. The nucleic acid of claim 22, wherein the polypeptide has a functional transmembrane domain.
- 28. The nucleic acid of claim 22, wherein the polypeptide is devoid of a functional transmembrane domain.
- 29. The nucleic acid of claim 22, wherein the polypeptide comprises SEQ ID NO:4.
- 30. The nucleic acid of claims 22 or 29 wherein the polypeptide has amino acids 431 to 454 deleted or inactivated.
- 31. The nucleic acid of claim 22, wherein the polypeptide is unglycosylated.
- 32. A method of using a nucleic acid molecule according to claim 22 comprising expressing said nucleic acid molecule in a cultured host cell transformed with a vector comprising said nucleic acid molecule operably linked to control sequences recognized by the host cell, and recovering the encoded polypeptide from the host cell.
- 33. A method for producing a polypeptide according to claim 22 comprising inserting into the DNA of a cell containing nucleic acid encoding said polypeptide a transcription regulatory element in sufficient proximity and orientation to the acid molecule to influence the transcription thereof, and inducibly or constitutively expressing the nucleic acid encoding said polypeptide using the transcription regulatory element.
