Claims
- 1. A method of producing a peptide having no L-methionine residue and having a molecular weight of 2,000 to 10,000, comprising;
- (A) cultivating prokaryotic cells carrying a recombinant pUC plasmid which contains a trp promoter and a DNA coding for a fused protein, said fused protein comprising;
- (1) a galK protein fragment comprising 200 to 300 amino acid residues from the N terminus of the galK protein, and
- (2) an L-methionine-free peptide having a molecular weight of 2,000 to 10,000 coupled to said galK protein fragment at the C terminus thereof via an L-methionine residue;
- to cause expression of said fused protein;
- (B) purifying said fused protein;
- (C) cleaving said purified fused protein with cyanogen bromide to yield a peptide mixture; and
- (D) isolating peptides which have a molecular weight of 2,000 to 10,000 from the peptide mixture to obtain said peptide having no L-methionine residue.
- 2. The method as claimed in claim 1, wherein the peptide having no L-methionine residue in the molecule and having a molecular weight of 2,000 to 10,000 has an amino acid sequence selected from the group consisting of:
- (a) H-His-Ser-Asp-Ala-Val-Phe-Thr-Asp-Asn-Tyr-Thr-Arg-Leu-Arg-Lys-Gln-Leu-Ala-Val-Lys-Lys-Tyr-Leu-Asn-Ser-Ile-Leu-Asn-Gly-Pro-Glu-Ala-OH (SEQ ID No: 1);
- (b) H-His-Ser-Asp-Ala-Val-Phe-Thr-Asp-Asn-Tyr-Thr-Arg-Leu-Arg-Lys-Gln-Leu-Ala-Val-Lys-Lys-Tyr-Leu-Asn-Ser-Ile-Leu-Asn-Gly-Pro-Glu-OH (SEQ ID No: 2); and
- (c) H-His-Ser-Asp-Ala-Val-Phe-Thr-Asp-Asn-Tyr-Thr-Arg-Leu-Arg-Lys-Gln-Leu-Ala-Val-Lys-Lys-Tyr-Leu-Asn-Ser-Ile-Leu-Asn-Gly-Pro-OH (SEQ ID No: 3).
- 3. The method as claimed in claim 1, wherein the expressing step (A) is effected by using a fed-batch fermentation method.
- 4. A recombinant pUC plasmid comprising a trp promoter and a DNA coding for a fused protein, said fused protein comprising:
- (1) a galK protein fragment comprising 200 to 300 amino acid residues from the N terminus of galK; and
- (2) an L-methionine-free peptide having a molecular weight of 2,000 to 10,000 coupled to said galK protein fragment at the C terminus thereof via an L-methionine residue.
