Claims
- 1. A method for expressing a nucleotide sequence of interest in a plant cell comprising,
a) providing an Agrobacterium replicon having a T-DNA, wherein said T-DNA comprises in a, 5′ to 3′ or a 3′ to 5′ orientation, a first target site for a site-specific recombinase, a viral replicon, said nucleotide sequence of interest operably linked to a first promoter that drives expression in said plant cell, and a second target site for said site-specific recombinase, wherein said first and said second target site are directly repeated and identical with respect to one another; b) infecting said plant cell with an Agrobacterium carrying said Agrobacterium replicon under conditions that allow transfer of said T-DNA and expression of said site-specific recombinase in said plant cell; and, wherein said plant cell, said T-DNA, or said viral replicon comprises a nucleotide sequence encoding said site-specific recombinase, an active fragment of said site-specific recombinase, or an active variant of said site-specific recombinase, wherein said nucleotide sequence is operably linked to a second promoter that drives expression in said plant cell, and wherein said active fragment or said active variant of said site-specific recombinase retains the ability to catalyze a recombination event.
- 2. The method of claim 1, wherein said site-specific recombinase is a member of the integrase family.
- 3. The method of claim 2, wherein said site-specific recombinase comprises FLP, Cre, Int, SSVI, Integrase R, or an active fragment or an active variant thereof.
- 4. The method of claim 3, wherein said first and said second target sites are selected from the group consisting of a FRT site and a mutant FRT site.
- 5. The method of claim 4, wherein said mutant FRT site is FRT5 (SEQ ID NO: 4), FRT6 (SEQ ID NO: 5) or FRT7 (SEQ ID NO: 6).
- 6. The method of claim 4, wherein said site-specific recombinase comprises an active variant of FLP.
- 7. The method of claim 4, wherein said site-specific recombinase comprises FLP.
- 8. The method of claim 4, wherein said site-specific recombinase comprises moFLP.
- 9. The method of claim 4, wherein said site-specific recombinase comprises an active fragment of FLP.
- 10. The method of claim 3, wherein said first and said second target sites are selected from the group consisting of a LOX site or a mutant LOX site.
- 11. The method of claim 10, wherein said site-specific recombinase comprises an active variant of Cre.
- 12. The method of claim 10, wherein said site-specific recombinase comprises Cre.
- 13. The method of claim 10, wherein said site-specific recombinase comprises moCre.
- 14. The method of claim 10, wherein said site-specific recombinase comprises an active fragment of Cre.
- 15. The method of claim 1, wherein said Agrobacterium replicon comprises a Ti plasmid.
- 16. The method of claim 1, wherein said Agrobacterium replicon comprises a Ri plasmid.
- 17. The method of claim 3, wherein said T-DNA comprises said nucleotide sequence encoding said site-specific recombinase, said active fragment of said site-specific recombinase, or said active variant of said site-specific recombinase.
- 18. The method of claim 3, wherein said viral replicon comprises said nucleotide sequence encoding said site-specific recombinase, said active fragment of said site-specific recombinase, or said active variant of said site-specific recombinase.
- 19. The method of claim 1, wherein said plant cell is from a monocot.
- 20. The method of claim 19, wherein said monocot is maize, wheat, rice, barley, sorghum, or rye.
- 21. The method of claim 20, wherein said monocot is maize.
- 22. The method of claim 1, wherein said plant cell is from a dicot.
- 23. The method of claim 22, wherein said dicot is soybean, canola, Brassica, sunflower, alfalfa, or safflower.
- 24. The method of claim 1, wherein said viral replicon is a MSV replicon, a WDV replicon, a TGMV replicon, or a TMV replicon.
- 25. The method of claim 1, wherein said second promoter is an inducible promoter.
- 26. The method of claim 1, wherein said plant cell is in a plant.
- 27. The method of claim 26, wherein said plant is a monocot.
- 28. The method of claim 27, wherein said monocot is maize, wheat, rice, barley, sorghum, or rye.
- 29. The method of claim 26, wherein said plant is a dicot.
- 30. A plant cell having a DNA construct comprising in a 5′ to 3′ or a 3′ to 5′ orientation a first target site for a first site-specific recombinase, a viral replicon, and a second target site for said first site-specific recombinase, wherein said first and said second target site are directly repeated and identical with respect to each other.
- 31. The plant cell of claim 30, wherein said DNA construct is stably incorporated into the genome of said plant cell.
- 32. The plant cell of claim 30, wherein said DNA construct comprises in a 5′ to 3′ or a 3′ to 5′ orientation said first target site for said first site-specific recombinase, said viral replicon, a nucleotide sequence of interest, and said second target site for said first site-specific recombinase.
- 33. The plant cell of claim 32, wherein said DNA construct is stably incorporated into the genome of said plant cell.
- 34. The plant of cell claim 33, wherein said nucleotide sequence of interest is operably linked to a promoter that drives expression in said plant cell.
- 35. The plant cell of claim 30, wherein said DNA construct is contained within a T-DNA.
- 36. The plant cell of claim 30, wherein said DNA construct further comprises a nucleotide sequence encoding said first site-specific recombinase.
- 37. The plant cell of claim 30, wherein said plant cell comprises a nucleotide sequence encoding said first site-specific recombinase.
- 38. The plant cell of claim 37, wherein said first site-specific recombinase is a member of the integrase family.
- 39. The plant cell of claim 38, wherein said first site-specific recombinase is FLP, Cre, Int, SSVI, Integrase R, or an active fragment or an active variant thereof.
- 40. The plant cell of claim 30, wherein said first and said second target site is an FRT site, a mutant FRT site, a LOX site, or a mutant LOX site.
- 41. The plant cell of claim 30, wherein said viral replicon is a MSV replicon, a WDV replicon, a TGMV replicon, or a TMV replicon.
- 42. The plant cell of claim 30, wherein said plant cell is from a monocot.
- 43. The plant cell of claim 42, wherein said monocot is maize, wheat, rice, barley, sorghum, or rye.
- 44. The plant cell of claim 43, wherein said monocot is maize.
- 45. The plant cell of claim 30, wherein said plant cell is from a dicot.
- 46. The plant cell of claim 45, wherein said dicot is soybean, canola, Brassica, sunflower, alfalfa, or safflower.
- 47 The plant cell of claim 30, wherein said plant cell is in a plant.
- 48. The plant cell of claim 47, wherein said plant is a monocot.

The plant cell of claim 32, wherein said DNA construct comprises in the 5′ to 3′ or the 3′ to 5′ orientation the first target site for said first site-specific recombinase, said viral replicon, a third target site for a second site-specific recombinase, the nucleotide sequence of interest, and the second target site for said first site-specific recombinase,
wherein said first and said second target site are directly repeated and identical with respect to each other, and said third target site is non-identical to said first and said second target site.
- 56. The plant cell of claim 55, wherein said DNA construct is stably incorporated into the genome of said plant cell.
- 57. The plant cell of claim 55, wherein said DNA construct is contained within a T-DNA.
- 58. The plant cell of claim 55, wherein said first and said second site-specific recombinase are identical.
- 59. The plant cell of claim 55, wherein said first or said second site-specific recombinase is FLP, Cre, Int, SSVI, or Integrase R, or an active fragment, or an active variant thereof.
- 60. The plant cell of claims 59, wherein said first, said second, or said third target sites are selected from the group consisting of a FRT site, a mutant FRT site, a LOX site, and a mutant LOX site.
- 61. The plant cell of claim 60, wherein said first or said second site-specific recombinase comprises FLP.
- 62. The plant cell of claim 60, wherein said mutant FRT site is a FRT5 site (SEQ ID NO:4), a FRT6 site (SEQ ID NO:5), or a FRT7 site (SEQ ID NO:6).
- 63. The plant cell of claim 60, wherein said first or said second site-specific recombinase comprises Cre.
- 64. The plant cell of claim 55, wherein said nucleotide sequence of interest is operably linked to a promoter that drives expression in said plant cell.
- 65. The plant cell of claim 55, wherein said DNA construct further comprises a nucleotide sequence encoding said first or said second site-specific recombinase.
- 66. The plant cell of claim 55, wherein said viral replicon is a MSV replicon, a WDV replicon, a TGMV replicon, or a TMV replicon.
- 67. The plant cell of claim 55, wherein said plant cell is from a monocot.
- 68. The plant cell of claim 67, wherein said monocot is maize, wheat, rice, barley, sorghum, or rye.
- 69. The plant cell of claim 68, wherein said monocot is maize.
- 70. The plant cell of claim 55, wherein said plant cell is from a dicot.
- 71. The plant cell of claim 70, wherein said dicot is soybean, canola, Brassica, sunflower, alfalfa or safflower.
