Claims
- 1. A genetic construct comprising
(1) a gene that encodes a fusion protein comprising (a) a first moiety which is a protein domain comprising an amino acid sequence foreign to filamentous phage and (b) a second moiety which comprises a mature filamentous phage coat protein; and (2) a promoter that is upstream of the gene and that regulates its transcription.
- 2. The genetic construct of claim 1 wherein the second moiety comprises a mature gene VIII protein of a filamentous phage.
- 3. The genetic construct of claim 1 wherein the second moiety comprises a mature gene III protein of a filamentous phage.
- 4. The genetic construct of any of claims 1 to 3 wherein the gene further encodes an amino terminal signal sequence.
- 5. The genetic construct of any of claims 1 to 3 wherein the first moiety is amino terminal to the second moiety.
- 6. The genetic construct of any of claims 1 to 3 wherein the fusion protein further comprises a peptide or amino acid linker disposed between the first and second moiety.
- 7. The genetic construct of any of claims 1 to 3 wherein the mature phage coat protein is a protein of M13 filamentous phage.
- 8. A library of nucleic acids that comprises a plurality of nucleic acid members that each comprise a first sequence encoding an antibody variable domain and a second sequence encoding a polypeptide comprising a mature gene VIII protein of a filamentous phage, wherein the nucleic acid members of the plurality differ by variegation of codons encoding amino acids of hypervariable regions of the antibody variable domain, and the first and second sequence encode components of a common fusion protein.
- 9. A library of nucleic acids that comprises a plurality of nucleic acid members that each comprise a first sequence encoding an antibody variable domain and a second sequence encoding a polypeptide comprising a mature gene III protein of a filamentous phage, wherein the nucleic acid members of the plurality differ by variegation of codons encoding amino acids of hypervariable regions of the antibody variable domain, and the first and second sequence encode components of a common fusion protein.
- 10. A nucleic acid encoding a fusion protein that comprises
(a) a first moiety that is a protein domain comprising an amino acid sequence foreign to a filamentous phage; and (b) a second moiety that comprises a mature filamentous phage coat protein.
- 11. The nucleic acid of claim 10 wherein the mature phage coat protein is encoded by gene VIII of filamentous phage.
- 12. The nucleic acid of claim 10 wherein the mature phage coat protein is encoded by gene III of filamentous phage.
- 13. The nucleic acid of any one of claims 10-12 wherein the first moiety comprises an antibody domain.
- 14. A genetic construct comprising
(1) a gene that encodes a fusion protein comprising (a) a first moiety which is a protein domain comprising an amino acid sequence foreign to filamentous phage and (b) a second moiety which comprises a functional domain of a filamentous phage coat protein; and (2) a promoter that is upstream of the gene and that regulates its transcription.
- 15. The construct of claim 14 wherein the phage coat protein of (b) is a gene III protein of a filamentous phage.
- 16. The construct of claim 14 wherein the phage coat protein of (b) is a gene VIII protein of a filamentous phage.
- 17. A library of nucleic acids that comprises a plurality of nucleic acid members that each comprise a first sequence encoding an antibody variable domain and a second sequence encoding a polypeptide comprising a functional domain of a coat protein of a filamentous phage, wherein the nucleic acid members of the plurality differ by variegation of codons encoding amino acids of hypervariable regions of the antibody variable domain, and the first and second sequence encode components of a common fusion protein.
- 18. The construct of claim 17 wherein the phage coat protein is a gene III protein of a filamentous phage.
- 19. The construct of claim 17 wherein the phage coat protein is a gene VIII protein of a filamentous phage.
- 20. A nucleic acid encoding a fusion protein that comprises
(a) a first moiety that is a protein domain comprising an amino acid sequence foreign to a filamentous phage; and (b) a second moiety that comprises a functional domain of a filamentous phage coat protein.
- 21. The nucleic acid of claim 20 wherein the functional domain is encoded by gene VIII of filamentous phage, or a fragment thereof.
- 22. The nucleic acid of claim 20 wherein the functional domain is encoded by gene III of filamentous phage, or a fragment thereof.
