Claims
- 1. A fusion product comprising a first DNA sequence coding for an amino acid sequence which is specifically cleaved by blood coagulation Factor X.sub.a and
- a second DNA sequence fused to said first DNA sequence, said second DNA sequence coding for a desired protein or peptide, the second DNA sequence being one which is not naturally fused to said first DNA sequence.
- 2. The product of claim 1, wherein said first DNA sequence codes for the amino acid sequence
- X--Y--Gly--Arg
- wherein X is selected from the group consisting of Ile, Leu, Pro and Ala, and Y is selected from the group consisting of Glu, Asp, Gln and Asn.
- 3. The product of claim 1, wherein said first DNA sequence codes for the amino acid sequence X--Y--Gly--Arg where X is Ile and Y is selected from the group consisting of Glu and Gln.
- 4. The product of claim 1, wherein said first DNA sequence codes for the amino acid sequence Ile--Glu--Gly--Arg.
- 5. The product of claim 1, containing the sequence
- ATC GAG GGT AGG.
- 6. The product of claim 5, containing the sequence
- ATC GAG GGT AGG CGT.

- 7. The product of claim 4, containing the sequence
- ATT GAA GGT CGT.
- 8.
- 8. A vector comprising a product according to claim 1 inserted therein.
- 9. A vector selected from the group consisting of M13mp11FX, M13mp11FX-MLC,
- pLcIIFX.beta., pLcIIFX-MLC, and
- pCT 20210.
- 10. The vector of claim 8, wherein said first DNA sequence coding for an amino acid sequence which is specifically cleaved by blood coagulation Factor Xa is also fused to an ATG codon.
- 11. The vector of claim 8, wherein said first DNA sequence coding for an amino acid sequence which is specifically cleaved by blood coagulation Factor Xa is also fused to a DNA sequence coding for at least part of a host protein.
- 12. The vector of claim 11, wherein the host protein is selected from the group consisting of lacZ, tufB, trpE, trpD, bla, cII and cat.
- 13. The vector of claim 8, wherein said first DNA sequence coding for an amino acid sequence which is specifically cleaved by blood coagulation Factor Xa is fused to the 5' end of the DNA sequence coding for said desired protein or peptide product.
- 14. The vector of claim 8, wherein said desired protein or peptide product is selected from the group consisting of an enzyme, a serum protein, a peptide hormone, and a precursor thereof.
- 15. The vector of claim 14, wherein said desired protein or peptide product is selected from the group consisting of .beta.-globin, calcitonin-glycine, and myosin light chain.
- 16. Host cells transformed with a vector according to claim 8.
- 17. Host cells transformed with a vector according to claim 9.
- 18. A process, for the production of a desired protein or peptide product in native form, comprising:
- transforming a host cell with a vector according to claim 8;
- expressing the desired protein or peptide product as a fusion protein comprising the desired protein or peptide product fused to the amino acid sequence which is specifically cleaved by blood coagulation Factor Xa; and
- cleaving the fusion protein with Factor Xa to yield the desired protein or peptide product in native form.
- 19. The process of claim 18, wherein the Factor Xa is prepared by activating Factor X.
- 20. The process of claim 18, wherein the Factor X is bovine Factor X.sub.1 or X.sub.2.
- 21. The process of claim 18, wherein the fusion protein further comprises at least a part of a host protein linked to the amino acid sequence which is specifically cleaved by blood coagulation Factor Xa.
- 22. The process of claim 21, including the step of purifying the fusion protein by use of an affinity chromatography medium specific for the host protein prior to the cleavage step.
- 23. The product of claim 1, further comprising a third DNA sequence coding for at least part of a host protein, said third DNA sequence fused to said first DNA sequence coding for an amino acid sequence which is specifically cleaved by a blood coagulation Factor Xa.
