Claims
- 1. An isolated nucleic acid encoding a hedgehog polypeptide having an amino acid sequence identical or homologous to a vertebrate hedgehog protein, or a portion thereof, and not identical to SEQ ID No. 34.
- 2. The nucleic acid of claim 1, which nucleic acid encodes a hedgehog polypeptide that modulates at least one of proliferation, differentiation or survival of mesodermal or ectodermal cells.
- 3. The nucleic acid of claim 1, which nucleic acid hybridizes under stringent conditions to a nucleic acid probe having a sequence represented by at least 12 consecutive nucleotides of a hedgehog nucleic acid selected from the group consisting of SEQ ID No:1, SEQ ID No:2, SEQ ID No:3, SEQ ID No:4, SEQ ID No:5, SEQ ID No:6 and SEQ ID No:7.
- 4. The nucleic acid of claim 1, further comprising a transcriptional regulatory sequence operably linked to said nucleotide sequence so as to render said nucleic acid suitable for use as an expression vector.
- 5. An expression vector, capable of replicating in at least one of a prokaryotic cell and eukaryotic cell, comprising the nucleic acid of claim 1.
- 6. A host cell transfected with the expression vector of claim 5 and expressing said hedgehog polypeptide.
- 7. A method of producing a recombinant hedgehog polypeptide comprising culturing the cell of claim 6 in a cell culture medium to express said hedgehog polypeptide and isolating said hedgehog polypeptide from said cell culture.
- 8. A nucleic acid composition comprising a substantially purified oligonucleotide, said oligonucleotide including a region of nucleotide sequence which hybridizes under stringent conditions to at least 25 consecutive nucleotides of sense or antisense sequence of SEQ ID No: 1, SEQ ID No:2, SEQ ID No:3, SEQ ID No:4, SEQ ID No:5, SEQ ID No:6, SEQ ID No:7, or naturally occurring mutants thereof.
- 9. The nucleic acid composition of claim 8, which oligonucleotide hybridizes under stringent conditions to at least 50 consecutive nucleotides of sense or antisense sequence of a nucleic acid selected from a group consisting of SEQ ID No: 1, SEQ ID No:2, SEQ ID No:3, SEQ ID No:4, SEQ ID No:5, SEQ ID No:6, SEQ ID No:7, or naturally occurring mutants thereof.
- 10. The nucleic acid composition of claim 8, which oligonucleotide further comprises a label group attached thereto and able to be detected.
- 11. The nucleic acid composition of claim 8, which oligonucleotide has at least one non-hydrolyzable bond between two adjacent nucleotide subunits.
- 12. A nucleic acid encoding a hedgehog polypeptide, which polypeptide comprises an amino acid sequence identical or homologous to an amino acid sequence designated in one of SEQ ID No:8, SEQ ID No:9, SEQ ID No:10, SEQ ID No:11, SEQ ID No:12, SEQ ID No:13 or SEQ ID No:14.
- 13. The nucleic acid of claim 12, which polypeptide comprises an amino acid sequence designated in SEQ ID No:40.
- 14. The nucleic acid of claim 12, which polypeptide is at least 70% homologous to a Sonic hedgehog (Shh) polypeptide.
- 15. The nucleic acid of claim 12, which polypeptide is at least 70% homologous to an Indian hedgehog (Ihh) polypeptide.
- 16. The nucleic acid of claim 12, which polypeptide is at least 70% homologous to a Desert hedgehog (Dhh) polypeptide.
- 17. The nucleic acid of claim 12, which polypeptide is a mammalian hedgehog polypeptide.
- 18. The nucleic acid of claim 12, which polypeptide antagonizes a biological activity of a wild-type hedgehog protein.
- 19. The nucleic acid of claim 12, which polypeptide mimics a biological activity of a wild-type hedgehog protein.
- 20. The nucleic acid of claim 12, which encoded polypeptide is a fusion protein.
- 21. A nucleic acid encoding a bioactive fragment of a vertebrate hedgehog polypeptide, which polypeptide has a molecular weight of approximately 19 kDa.
- 22. The nucleic acid of claim 21, which polypeptide comprises an amino acid sequence identical or homologous to an amino acid sequence designated in one of SEQ ID No:8, SEQ ID No:9, SEQ ID No:10, SEQ ID No:11, SEQ ID No:12, SEQ ID No: 13 or SEQ ID No:14.
- 23. A nucleic acid encoding a polypeptide, which polypeptide comprises, as a hedgehog portion, an amino acid sequence represented by the formula A-B wherein,
A represents all or a portion of the amino acid sequence designated by residues 1-168 of SEQ ID No:40; and B represents at least one amino acid residue of the amino acid sequence designated by residues 169-221 of SEQ ID No:40; wherein A and B together represent a contiguous polypeptide sequence represented by SEQ ID No:40, and said polypeptide possess a biochemical activity of a hedgehog protein.
- 24. The nucleic acid of claim 23, wherein B represents at least five amino acid residues of the amino acid sequence designated by residues 169-221 of SEQ ID No:40.
- 25. The nucleic acid of claim 23, wherein B represents at least ten amino acid residues of the amino acid sequence designated by residues 169-221 of SEQ ID No:40.
- 26. The nucleic acid of claim 23, which polypeptide is a fusion protein.
- 27. The nucleic acid of claim 23, which polypeptide agonizes the biological activity of a wild-type sonic hedgehog (Shh) protein.
- 28. The nucleic acid of claim 23, which polypeptide antagonizes the biological activity of a wild-type sonic hedgehog (Shh) protein.
- 29. A nucleic acid encoding a polypeptide, which polypeptide comprises, as a hedgehog portion, an amino acid sequence represented by the formula A-B wherein,
A represents all or the portion of the amino acid sequence designated by residues 24-193 of SEQ ID No: 13; and B represents at least one amino acid residue of the amino acid sequence designated by residues 194-250 of SEQ ID No:13; wherein A and B together represent a contiguous polypeptide sequence designated in SEQ ID No: 13, and said polypeptide possess a biochemical activity of a hedgehog protein.
- 30. A nucleic acid encoding a polypeptide, which polypeptide comprises, as a hedgehog portion, an amino acid sequence represented by the formula A-B wherein,
A represents all or the portion of the amino acid sequence designated by residues 25-193, or analogous residues thereof, of a vertebrate hedgehog polypeptide identical or homologous to SEQ ID No: l 1; and B represents at least one amino acid residue of the amino acid sequence designated by residues 194-250, or analogous residues thereof, of a vertebrate hedgehog polypeptide identical or homologous to SEQ ID No: 11; wherein A and B together represent a contiguous polypeptide sequence identical or homologous to SEQ ID No: 11, and said polypeptide possess a biochemical activity of a hedgehog protein.
- 31. A nucleic acid encoding a polypeptide, which polypeptide comprises, as a hedgehog portion, an amino acid sequence represented by the formula A-B wherein,
A represents all or the portion of the amino acid sequence designated by residues 23-193, or analogous residues thereof, of a vertebrate hedgehog polypeptide identical or homologous to SEQ ID No:9; and B represents at least one amino acid residue of the amino acid sequence designated by residues 194-250, or analogous residues thereof, of a vertebrate hedgehog polypeptide identical or homologous to SEQ ID No:9; wherein A and B together represent a contiguous polypeptide sequence designated in SEQ ID No:9, and said polypeptide possess a biochemical activity of a hedgehog protein.
- 32. A nucleic acid encoding a polypeptide, which polypeptide comprises, as a hedgehog portion, an amino acid sequence represented by the formula A-B wherein,
A represents all or the portion of the amino acid sequence designated by residues 28-197, or analogous residues thereof, of a vertebrate hedgehog polypeptide identical or homologous to SEQ ID No:10; and B represents at least one amino acid residue of the amino acid sequence designated by residues 198-250, or analogous residues thereof, of a vertebrate hedgehog polypeptide identical or homologous to SEQ ID No:10; wherein A and B together represent a contiguous polypeptide sequence identical or homologous to SEQ ID No:10, and said polypeptide possess a biochemical activity of a hedgehog protein.
- 33. A nucleic acid encoding a polypeptide, which polypeptide comprises, as a hedgehog portion, an amino acid sequence represented by the formula A-B wherein,
A represents all or the portion of the amino acid sequence designated by residues 1-98 of SEQ ID No:14; and B represents at least one amino acid residue of the amino acid sequence designated by residues 99-150 of SEQ ID No:14; wherein A and B together represent a contiguous polypeptide sequence designated in SEQ ID No: 14, and said polypeptide possess a biochemical activity of a hedgehog protein.
- 34. A nucleic acid which hybridizes under stringent conditions to a portion of the nucleotide sequence designated by one of SEQ ID No:1-7 and specifically detects a vertebrate hedgehog gene or mRNA.
