Claims
- 1. A transgene construct, comprising a transgene encoding a polypeptide comprising a rod outer segment (ROS) targeting signal, and an operably associated rod-specific regulatory sequence, wherein said polypeptide is not rhodopsin.
- 2. The construct of claim 1, wherein said polypeptide is a G protein-coupled receptor (GPCR).
- 3. The construct of claim 2, wherein said GPCR is a cannabinoid receptor.
- 4. The construct of claim 1, wherein said polypeptide is a fusion protein.
- 5. The construct of claim 1, wherein said ROS targeting signal comprises SEQ ID NO:4.
- 6. The construct of claim 1, wherein said rod-specific regulatory sequence comprises a rhodopsin promoter.
- 7. A vector comprising the construct of claim 1.
- 8. A cell comprising the construct of claim 1.
- 9. A mouse cell whose genome comprises a transgene encoding a polypeptide comprising a ROS targeting signal operably associated with a rod-specific regulatory sequence, wherein said polypeptide is not a rhodopsin.
- 10. The mouse cell of claim 9, further comprising a functional disruption of one or both endogenous rhodopsin gene alleles.
- 11. The mouse cell of claim 9, wherein said polypeptide is a GPCR.
- 12. The mouse cell of claim 11, wherein said GPCR is a cannabinoid receptor.
- 13. The mouse cell of claim 9, wherein said polypeptide is a fusion protein.
- 14. The mouse cell of claim 9, wherein said ROS targeting signal comprises SEQ ID NO:4.
- 15. The mouse cell of claim 10, wherein said genome comprises a functional disruption of both endogenous rhodopsin gene alleles.
- 16. The mouse cell of claim 9, further comprising an embryonic stem cell.
- 17. The mouse cell of claim 9, further comprising a rod cell.
- 18. An extract of the mouse cell of claim 9, comprising an outer segment membrane of said cell.
- 19. A mouse whose genome comprises a transgene encoding a polypeptide comprising a ROS targeting signal operably associated with a rod-specific regulatory sequence, wherein said polypeptide is not a rhodopsin.
- 20. The mouse of claim 19, further comprising a functional disruption of one or both endogenous rhodopsin gene alleles.
- 21. The mouse of claim 19, wherein said polypeptide is a GPCR.
- 22. The mouse of claim 21, wherein said GPCR is a cannabinoid receptor.
- 23. The mouse of claim 19, wherein said polypeptide is a fusion protein.
- 24. The mouse of claim 19, wherein said ROS targeting signal comprises SEQ ID NO:4.
- 25. The mouse of claim 20, wherein said genome comprises a functional disruption of both endogenous rhodopsin gene alleles.
- 26. A rod cell or outer membrane extract thereof isolated from the mouse of claim 19.
