Claims
- 1. A hemoglobin-like protein bearing prosthetic heme groups and having reversible oxygen-binding activity, said protein comprising at least one polypeptide which is a non-naturally occurring, genetically fused globin-like polypeptide comprising two or more globin-like domains connected by one or more peptide bonds into a single unbranched polypeptide chain, said chain being capable of associating with alpha and/or beta globin and incorporating heme to form a hemoglobin-like protein with reversible oxygen binding activity.
- 2. The protein of claim 1 said polypeptide comprising only two globin-like domains.
- 3. The protein of claim 2 said protein being a di-alpha globin-like polypeptide comprising first and second alpha globin-like polypeptide domains, said chain being capable of associating with beta globin.
- 4. The protein of claim 2, said protein being a di-beta globin-like polypeptide comprising first and second beta-globin-like polypeptide domains, said chain being capable of associating with alpha-globin activity.
- 5. The protein of claim 2, said protein being a pseudodimeric globin-like polypeptide comprising an alpha globin-like polypeptide domain and a beta globin-like polypeptide domain.
- 6. The protein of claim 1 wherein in said polypeptide a first and a second globin-like polypeptide domain are connected by a peptide linker of one or more amino acids.
- 7. The protein of claim 6 wherein the amino acids of the linker in said polypeptide are selected from the group consisting of lysine, aspartic acid, arginine, serine, asparagine, proline and glycine.
- 8. The protein of claim 7 wherein the linker in said polypeptide consists of one or more glycines.
- 9. The protein of claim 1 wherein in said polypeptide at least one globin-like domain has at least 75% sequence identity with human alpha globin or human beta globin.
- 10. The protein of claim 1 wherein in said polypeptide each globin-like domain is identical in sequence with human alpha globin or human beta globin.
- 11. The protein of claim 1 wherein at a predetermined position, a first globin-like domain has a cysteine residue and a second globin-like domain has a residue other than cysteine.
- 12. The protein of claim 11 wherein one of said predetermined positions is
- (a) a position in the alpha globin-like domain corresponding to a human alpha globin position selected from the group consisting of his72, asn78, asn68, ala71, thr67, lys7, lys11, thr8, ala12, thr118, lys16, ala45, glu116, gly15, his112, thr24, glu23, lys60, lys56, his50, gly51, glu53, ser49, asp47, gln54, his45, lys90, ala82, lys61, ala19, his20, asp85, ser81, asp75, asp74, lys139, asp64, and gly18, or
- (b) a position in a beta globin-like domain corresponding to a human beta globin position selected from the group consisting of asp79, his2, leu3, thr4, glu6, ser9, thr12, ala13, gly16, lys17, val18, asn19, val20, asp21, glu22, lys65, ser72, ala76, his77, asp79, asn80, gly83, ala86, thr87, glu90, lys95, lys59, glu43, ser44, asp47, ser49, thr50, ala53, asp52, lys61, glu121, lys120, thr123, lys66, asp73, ala62, his116, and his117.
- 13. The protein of claim 1, said protein being a pseudotetramer.
- 14. The protein of claim 1, said protein being a pseudooctamer composed of two pseudotetramers, or of one pseudotetramer and one tetramer.
- 15. The protein of claim 14, said protein being
- (a) a polypeptide comprising four alpha globin-like domains, and, in the form of one or more additional polypeptides, four associable beta globin-like domains, or
- (b) a polypeptide comprising four beta globin-like domains, and, in the form of one or more additional polypeptides, four associable alpha globin-like domains.
- 16. The protein of claim 15, wherein the associable domains are domains of pseudodimeric globin-like polypeptides.
- 17. The protein of claim 15 wherein the associable domains are each provided as a separate polypeptide chain.
- 18. A human hemoglobin-like protein according to claim 1, said protein comprising:
- (a) a di-alpha globin-like polypeptide and two beta-globin-like polypeptides;
- (b) a di-beta-globin-like polypeptide and two alpha globin-like polypeptides;
- (c) a di-alpha globin-like polypeptide and a di-beta globin-like polypeptide;
- (d) an alpha/beta di-globin-like polypeptide, an alpha-globin like polypeptide, and a beta globin-like polypeptide, or;
- (e) two alpha/beta digloblin-like poly-peptides.
- 19. The protein of claim 18, said protein being a pseudotetramer.
- 20. The protein of claim 19, said protein comprising a di-alpha-globin-like polypeptide and two beta-globin-like polypeptides.
- 21. The protein of claim 20, wherein the alpha globin-like domains of said di-alpha-globin-like polypeptide each have at least 75% sequence identity with human alpha globin, and the beta globin-like polypeptides each have at least 75% sequence identity with human beta globin.
- 22. The protein of claim 21, wherein the alpha globin-like domains are identical in sequence with human alpha globin and the beta globin-like polypeptides are identical in sequence with human beta globin.
- 23. The protein of claim 21 which is a low affinity hemoglobin mutant.
- 24. The protein of claim 21 which is a high affinity hemoglobin mutant.
- 25. The protein of claim 21 wherein the beta globin-like polypeptide comprises a Presbyterian mutation.
- 26. The protein of claim 18, said protein being a pseudooctamer composed of two pseudotetramers, or of one pseudotetramer and one tetramer.
- 27. The protein of claim 26, said protein comprising two covalently bonded pseudotetramers, each pseudotetramer comprising (a) a pseudodimer selected from the group consisting of a di-alpha-globin-like polypeptide, (b) a di-beta-globin-like polypeptide, and (c) an alpha/beta globin-like polypeptide, the pseudodimers of each pseudotetramer being covalently bonded, said pseudodimers being asymmetric, said asymmetry being the presence at a amino acid position of a cysteine in a first domain but not in a second domain.
- 28. The protein of claim 27 wherein said pseudodimers are covalently bonded by a disulfide bridge between a cysteine of one psuedodimer and a cysteine of the other psuedodimer.
- 29. The protein of claim 27 wherein said pseudodimers are covalently bonded by a bridging moiety, other than a disulfide bridge, between a cysteine of one pseudodimer and a cysteine of the other pseudodimer.
- 30. The protein of claim 29 wherein the bridging moiety is nonreducible under physiologic conditions.
- 31. The protein of claim 29 wherein the bridging moiety increases the intravascular retention time of the protein.
- 32. The protein of claim 29 wherein the bridging moiety reduces the isoelectric point of the protein.
- 33. The protein of claim 26, said protein comprising a single unbranched polypeptide comprising more than two alpha globin-like domains or more than two beta globin-like domains.
- 34. The protein of claim 18, said protein comprising more than two pseudotetramers.
- 35. The protein of claim 34, each pseudotetramer comprising (a) a pseudodimer selected from the group consisting of a di-alpha-globin-like polypeptide, (b) a di-beta-globin-like polypeptide, and (c) an alpha/beta globin-like polypeptide, the pseudodimers of each pseudotetramer being covalently bonded, said pseudodimers being asymmetric, said asymmetry being the presence at a amino acid position of a cysteine in a first domain but not in a second domain.
- 36. The protein of claim 35, said pseudotetramers being covalently bonded to a coiled coil linker means.
- 37. The protein of claim 35, said psuedotetramers being covalently bonded to a multihelical bundle linker means.
- 38. The protein of claim 1 wherein said hemoglobin-like protein is a low oxygen affinity mutant hemoglobin.
- 39. The protein of claim 38 wherein at least one of said low affinity mutations is

- 45. The polypeptide of claim 44 wherein said globin-like domains of said polypeptide are identical in sequence, except that at one or more predetermined positions, a first globin-like domain has a cysteine residue and a second globin-like domain has a residue other than cysteine.
- 46. The protein of claim 1 wherein each alpha globin-like domain of said genetically fused polypeptide is identical to human alpha globin save for
- (a) one or more an oxygen affinity-modifying modifications characterized as

- 48. The protein of claim 1, said protein comprising more than two pseudotetramers.
- 49. An octameric hemoglobin-like protein, wherein (a) the two beta-globin-like polypeptides in the beta.sub.1 position are disulfide bonded and the two beta-globin-like polypeptides in the beta.sub.2 position are disulfide bonded, or (b) the two alpha-globin-like polypeptides in the alpha.sub.1 position are disulfide bonded and the two alpha-globin-like polypeptides in the alpha.sub.2 position are disulfide bonded.
- 50. A genetically fused hemoglobin-like protein composed of fewer than four polypeptide chains and having a half-life in the circulatory system substantially in excess of that for des-Val hemoglobin.
