Claims
- 1. A method for inhibiting telomerase activity in human cells, comprising transferring into cells an exogenous polynucleotide comprising a transcriptional unit having a polynucleotide sequence of at least 25 consecutive nucleotides which is substantially identical or substantially complementary to a human telomerase RNA sequence operably linked to a heterologous transcriptional regulatory sequence which promotes transcription of operably linked polynucleotides in said cells.
- 2. The method of claim 1, wherein the cells are neoplastic cells.
- 3. The method of claim 1, wherein the exogenous polynucleotide comprises the sequence:

- 4. The method of claim 1, wherein the heterologous transcriptional regulatory sequence comprises a promoter which is constitutively active in human cells.
- 5. The method of claim 1, wherein the exogenous polynucelotide is an adenoviral genome having a hTR transcription unit.
- 6. The method of claim 1, wherein said transcription unit produces antisense RNA substantially complementary to hTR RNA.
- 7. The method of claim 1, wherein the exogenous polynucleotide comprises the sequence:

A polynucleotide for gene therapy of a human disease, comprising a transcription unit comprising a polynucleotide sequence of at least 25 consecutive nucleotides which is substantially identical or substantially complementary to a human telomerase RNA sequence operably linked to a heterologous transcriptional regulatory sequence which promotes transcription of operably linked polynucleotides in said cells.
- 9. A ribozyme which cleaves hTR or human telomeric repeat sequences.
