Claims
- 1. An oligonucleotide in substantially pure form that is 10 to 500 nucleotides in length and that comprises 10 to 500 nucleotides in a sequence identical or exactly complementary to a contiguous sequence of human telomerase RNA component, the oligonucleotide characterized by the ability to specifically hybridize in a cell or tissue sample or extract only to human telomerase RNA component or its complement, wherein the oligonucleotide does not hybridize to telomeric DNA.
- 2. The oligonucleotide of claim 1 that is an oligodeoxyribonucleotide.
- 3. The oligonucleotide of claim 1 that is an oligoribonucleotide.
- 4. The oligonucleotide of claim 1 that, when bound to an RNA component of human telomeras inhibits or blocks the activity of the telomerase.
- 5. A recombinant expression plasmid comprising a nucleotide sequence identical or exactly complementary to a contiguous sequence 10 to 500 nucleotides in length of human telomerase RNA component and further comprising a promoter positioned to drive transcription of an RNA complementary or identical in sequence to the nucleotide sequence, wherein the RNA specifically hybridizes only to human telomerase RNA component or its complement, and does not hybridize to telomeric DNA.
- 6. The recombinant expression plasmid of claim 5, wherein said plasmid functions to produce the oligonucleotide in eukaryotic host cell.
- 7. The recombinant expression plasmid of claim 5, wherein said plasmid functions to produce the oligonucleotide in prokaryotic host cells.
- 8. An RNA component of a mammalian telomerase in substantially pure form that comprises a sequence that is substantially homologous to a sequence in the RNA component of human telomerase.
- 9. A method for producing a recombinant telomerase enzyme, said method comprising transforming a eukaryotic host cell that expresses protein components of telomerase with a recombinant expression vector that encodes an RNA component of claim 8, and culturing said host cells transformed with said vector under conditions such that the protein components and RNA component are expressed and assemble to form an active telomerase molecule capable of adding sequences to telomeres of chromosomal DNA.
- 10. The oligonuclcolide of claim 1 wherein the sequence is identical or exactly complementary to a contiguous sequence contained within a human genomic DNA sequence encoding the RNA component of human telomerase located in an .about.2.5 kb HindIII-SacI insert of plasmid pGRN33 (ATCC 75926).
- 11. The oligonucleotide of claim 2 that is 5'- CTCAGTTAGG GTTAGACAAA -3' (SEQ ID NO:26); 5'- CGCCCTTCTC AGTTAGGGTTA G -3' (SEQ ID NO:27); 5'- GGCGCCTACG CCCTTCTCAG TT -3' (SEQ ID NO:28); or 5'- CAGGCCCACC CTCCGCAACC -3' (SEQ ID NO:7).
- 12. The oligonucleotide of claim 3 that is 5'- CUCAGUUAGG GUUAGACAAA -3' (SEQ ID NO:4); 5'- CGCCCUUCUC AGUUAGGGUUA G -3' (SEQ ID NO:5); 5'- GGCGCCUACG CCCUUCUCAG UU -3'(SEQ ID NO:6); or 5'- CAGGCCCACC CUCCGCAACC -3' (SEQ ID NO:29).
- 13. An oligonucleotide of 10 to 30 nucleotides in length in a sequence identical or exactly complementary to a contiguous sequence of human telomerase RNA component, wherein the oligonucleotide is hybridized, in a cell or tissue sample or extract, only to a nucleic acid having the sequence of a mammalian telomerase RNA component or its complement.
- 14. The oligonucleotide of claim 13 that comprises a modified nucleotide analogue.
- 15. The oligonucleotide of claim 13 that is a heteropolymer comprising deoxyribonucleotides, ribonucleotides or an analogue of either.
- 16. The oligonucleotide of claim 13 that is identical in sequence to the contiguous sequence.
- 17. The oligonucleotide of claim 13 that is complementary in sequence to the contiguous sequence.
- 18. The oligonucleotide of claim 13 further comprising a label.
- 19. The oligonucleotide of claim 13 wherein the mammalian telomerase is human telomerase.
- 20. The oligonucleotide of claim 14 wherein the modified nucleotide analogue is a phosphorothioate nucleotide, a chiral-methyl phosphonate nucleotide or a 2'-O-methyl ribonucleotide.
- 21. The oligonucleotide of claim 16 comprising the sequence 5'-CCCTAA -3', wherein "T" also can be "U".
- 22. The oligonucleotide of claim 16 that does not comprise the sequence 5'- CCCTAA -3', wherein "T" also can be "U".
- 23. The oligonucleotide of claim 17 comprising the sequence 5'-TTAGGG -3', wherein "T" also can be "U".
- 24. The oligonucleotide of claim 17 that does not comprise the sequence 5'- TTAGGG -3', wherein "T" also can be "U".
- 25. The oligonticleotide of claim 18 wherein the label is a radioactive label, a fluorescent label, biotin or avidin.
- 26. The recombinant expression plasmid of claim 5 wherein the RNA comprises the sequence 5'- CCCUAA -3'.
- 27. The recombinant expression plasmid of claim 5 wherein the RNA does not comprise the sequence 5'- CCCUAA -3'.
- 28. The recombinant expression plasmid of claim 5 wherein the RNA comprises the sequence 5'- UUAGGG -3'.
- 29. The recombinant expression plasmid of claim 5 wherein the RNA does not comprise the sequence 5'- UUAGGG -3'.
- 30. A host cell transformed with a recombinant expression plasmid comprising a nucleotide sequence identical or exactly complementary to a contiguous sequence 10 to 500 nucleotides in length of human telomerase RNA component, and further comprising a promoter positioned to drive transcription of an RNA complementary or identical in sequence to the nucleotide sequence, wherein the RNA specifically hybridizes only to human telomerase RNA component or its complement and does not hybridize to telomeric DNA.
- 31. The host cell of claim 30 that is a eukaryotic cell.
- 32. The host cell of claim 30 that is a prokaryotic cell.
- 33. The host cell of claim 30 wherein the contiguous sequence is contained within a human genomic DNA sequence encoding the RNA component of human telomerase located in an .about.2.5 kb HindIII-SacI insert of plasmid pGRN33 (ATCC 75926).
- 34. An oligonucleotide probe or primer 10 to 30 nucleotides in length that specifically binds onlv to human telomerase RNA component or to either strand of a gene encoding human telomerase RNA component, wherein the oligonucleotide does not hybridize to telomeric DNA.
- 35. The oligonucleotide of claim 34 that is a primer that initiates DNA synthesis in a polymerization reaction.
- 36. The oligonucleotide of claim 34 that is a probe.
- 37. The oligonucleotide of claim 34 that specifically binds to the RNA component of human telomerase.
- 38. The oligonucleotide of claim 34 that is hybridized in a cell or tissue sample or extract to a nucleic acid comprising the sequence of human telomerase RNA or its complement.
- 39. The oligonucleotide of claim 34 that specifically binds to a contiguous sequence contained within a human genomic DNA sequence encoding human telomerase RNA component located in an .about.2.5 kb HindIII-SacI insert of plasmid pGRN33 (ATCC 75926).
- 40. The oligonucleotide primer of claim 35 that is 5'- GGAAGGTCTG AGACTAG -3' (SEQ ID NO:20) or 5'- ATCTCCTGCC CAGTCTG -3' (SEQ ID NO:21), wherein "T" also can be "U".
- 41. The oligonucleotide probe of claim 36 further comprising a label.
- 42. The oligonucleotide of claim 38 hybridized to a cell or tissue sample in situ.
- 43. The oligonucleotide probe of claim 41 wherein the label is a radioactive label, a fluorescent label, biotin or avidin.
- 44. The oligonucleotide of claim 1 comprising a label.
- 45. The oligonucleotide of claim 13 that is an oligodeoxyribonucleotide.
- 46. The obigonucleotide of claim 13 that is an oligoribonucleotide.
- 47. The oligonucleotide of claim 34 that is an oligodeoxyribonucleotide.
- 48. The oligonucleotide of claim 34 that is an oligoribonucleotide.
