Claims
- 1. An oligonucleotide which specifically binds with the promoter region of rat .alpha.1(I) collagen gene to form a triplex to block expression of the collagen gene in cell culture.
- 2. The oligonucleotide of claim 1, said oligonucleotide binding to a polypurine-polypyrimidine region of said promoter region.
- 3. The oligonucleotide of claim 2, said polypurine-polypyrimidine region having at least a part of the sequence of SEQ ID No. 1.
- 4. The oligonucleotide of claim 1, said oligonucleotide comprising the sequence selected from the group consisting of SEQ ID Nos. 5, 6, 8, 9, 11 and 12.
- 5. The oligonucleotide of claim 1, said oligonucleotide binding in an anti-parallel orientation with said promoter region.
- 6. The oligonucleotide of claim 1, said oligonucleotide having a length of from about 5-50 nucleotides.
- 7. The oligonucleotide selected from the group consisting of SEQ ID Nos. 5, 6, 8, 9, 11 and 12.
- 8. An olignonucleotide which specifically binds with rat collagen .alpha.1(I) promoter to form a triplex block the expression of a gene operably linked to the promoter in rat cell culture.
- 9. The oligonucleotide of claim 8, said oligonucleotide binding to a polypurine-polypyrimidine region of said promoter region.
- 10. The oligonucleotide of claim 9, said polypurine-polypyrimidine region having at least a part of the sequence of SEQ ID No. 1.
- 11. The oligonucleotide of claim 8, said oligonucleotide comprising the sequence selected from the group consisting of SEQ ID Nos. 5, 6, 8, 9, 11 and 12.
- 12. The oligonucleotide of claim 8, said oligonucleotide binding in an anti-parallel orientation with said promoter region.
- 13. The oligonucleotide of claim 8, said oligonucleotide having a length of from about 5-50 nucleotides.
