Claims
- 1. An isolated polynucleotide from the gene for human telomerase reverse transcriptase (hTRT) comprising a sequence having at least one of the following features:
a) it contains at least 25 consecutive nucleotides within SEQ. ID NO:6 but not within SEQ. ID NO:1. b) it is complementary to a); or c) it contains at least 25 consecutive nucleotides within the insert of Lambda phage GΦ5 (ATCC Accession No. 98505), but not within the insert of Lambda phage 25-1.1 (ATCC Accession No. 209024).
- 2. The polynucleotide of claim 1, wherein said sequence contains at least 25 consecutive nucleotides within SEQ. ID NO:6 but not within SEQ. ID NO: 1, or the complement thereof.
- 3. The polynucleotide of claim 1, wherein said sequence contains at least 50 consecutive nucleotides within SEQ. ID NO:6 but not within SEQ. ID NO: 1, or the complement thereof.
- 4. The polynucleotide of claim 1, wherein said sequence contains at least 100 consecutive nucleotides within SEQ. ID NO:6 but not within SEQ. ID NO:1, or the complement thereof.
- 5. The polynucleotide of claim 1, wherein said sequence contains at least 200 consecutive nucleotides within SEQ. ID NO:6 but not within SEQ. ID NO: 1, or the complement thereof.
- 6. The polynucleotide of claim 1, wherein said sequence contains at least 500 consecutive nucleotides within SEQ. ID NO:6 but not within SEQ. ID NO: 1, or the complement thereof.
- 7. The polynucleotide of claim 1, wherein said sequence contains at least 25 consecutive nucleotides within the BglII-FSPI fragment in the insert of Lambda phage GΦ5, but not within the insert of Lambda phage 25-1.1.
- 8. The polynucleotide of claim 7, wherein said sequence contains at least 50 consecutive nucleotides within said fragment.
- 9. The polynucleotide of claim 7, wherein said sequence contains at least 100 consecutive nucleotides within said fragment.
- 10. The polynucleotide of claim 1, wherein said sequence contains at least 200 consecutive nucleotides within said fragment.
- 11. The polynucleotide of claim 1, wherein said sequence contains at least 500 consecutive nucleotides within said fragment.
- 12. The polynucleotide of claim 1, wherein said sequence is within 1.8 kilobases upstream of the translation initiation site.
- 13. The polynucleotide of claim 3, wherein said sequence is within 1.8 kilobases upstream of the translation initiation site.
- 14. The polynucleotide of claim 8, wherein said sequence is within 1.8 kilobases upstream of the translation initiation site.
- 15. The polynucleotide of claim 1, which promotes transcription in cells endogenously expressing hTRT.
- 16. A method of gene expression, comprising expressing a heterologous gene in a cell under control of the polynucleotide of claim 1.
- 17. The method of claim 16, wherein the heterologous gene is a reporter gene.
- 18. The method of claim 17, wherein the reporter gene is luciferase, glucuronidase, galactosidase, chloramphenicol acetyl transferase, green fluorescent protein (GFP) or alkaline phosphatase.
- 19. The method of claim 16, conducted in order to measure a cis or trans transcriptional regulatory factor that regulates transcription of hTRT.
- 20. The method of claim 16, conducted for the purpose of drug screening.
